目的 :观察补肾益智方对AD动物模型大脑神经元磷酸化Tau蛋白及其相关酶类蛋白磷酸酶 1(PP 1)、蛋白磷酸酶 2A(PP 2A)的影响。方法 :用Wistar大鼠 80只以D 半乳糖腹腔注射 4周加上鹅膏覃酸 (ibotenicacid ,IBO)脑内Meynert核注射制造AD模型 ,随机分成AD模型组、双益平对照组、补肾益智方高、低剂量治疗组加上正常老年组和正常青年组总共设立六个组别。动物经治疗处理 6周后 ,应用免疫组织化学方法检测大鼠海马CA1和齿状回磷酸化Tau蛋白、PP 1、PP 2A神经元数目和光密度。结果 :AD模型组海马内神经元的Tau蛋白过度磷酸化而相互集结 ,形成成对螺旋细丝 ,表达脱Tau蛋白磷酸化的磷酸酯酶PP 1、PP 2A均明显减少 ;而补肾益智方治疗组可以使这种现象得到明显改善。结论 :补肾益智方能较有效地改善AD大鼠脑内神经元的Tau蛋白过度磷酸化 ,促进PP 1、PP 2A的表达。
Objective : To observe the effect of Bushen Yizhi Recipe on phosphorylated Tau protein and its related enzyme-like protein phosphatase 1 (PP 1) and protein phosphatase 2A (PP 2A) in neurons of AD animal models. METHODS: AD models were made by 80 intraperitoneal injections of D-galactose in 80 Wistar rats and intrathecal injection of Meynert for 4 weeks. They were randomly divided into AD model group, Shuangyi Ping control group, Bushenyi A total of six groups were set up in the high-, low-dose treatment group plus the normal elderly group and the normal youth group. Six weeks after treatment, animals were examined for the number of neurons and optical density of hippocampal CA1 and dentate gyrophosphorylated Tau protein, PP 1 and PP 2A by immunohistochemistry. Results: In the AD model group, the Tau protein in the hippocampal neurons was hyperphosphorylated and aggregated to form a pair of spiral filaments. The expression of dephosphorylated phospholipase PP 1 and PP 2A were significantly reduced in the AD model group. Treatment groups can significantly improve this phenomenon. Conclusion: Bushen Yizhi Recipe can effectively improve the hyperphosphorylation of Tau protein in neurons of AD rats and promote the expression of PP 1 and PP 2A.
