目的:观察小柴胡汤含药血清对SMMC-7721人肝癌细胞是否具有分化诱导作用,并探讨其分化诱导的作用机制。方法:经药物作用后,观察细胞形态、MTT抑制率、乳酸脱氢酶(LDH)、琥珀酸脱氢酶(SDH)、细胞周期的情况。结果:小柴胡汤含药血清使SSMC-7721细胞株有向成熟细胞形态分化的趋势;MTT抑制作用增强;细胞内LDH活性降低、SDH活性上升;流式测定S期细胞数减少,细胞周期阻滞于G0/G1期。其中以低剂量组效果最为显著(P<0.05)。结论:小柴胡汤含药血清可诱导SSMC-7721细胞株向成熟细胞分化,其作用机制可能与降低细胞内LDH活性、升高SDH活性,并且将细胞阻滞于G0/G1期有关。
OBJECTIVE: To observe whether Xiao Chai Hu Tang serum contains differentiation-inducing effect on SMMC-7721 human hepatocarcinoma cells and to explore the mechanism of its differentiation-induced effect. Methods: The cell morphology, MTT inhibition rate, lactate dehydrogenase (LDH), succinate dehydrogenase (SDH) and cell cycle were observed after drug treatment. Results: The drug-containing serum from Xiao Chai Hu Tang showed the tendency of differentiation towards mature cells in SSMC-7721 cells; the inhibition of MTT increased; the activity of LDH decreased and the activity of SDH increased; the number of cells in S phase decreased and the cell cycle resistance Lag in G0 / G1 period. Among them, the effect of low dose group was the most significant (P <0.05). CONCLUSION: Xiao Chai Hu Tang containing serum can induce SSMC-7721 cell line to differentiate into mature cells. Its mechanism may be related to decreasing intracellular LDH activity, increasing SDH activity, and arresting cells in G0 / G1 phase.
