目的:研究分析乙肝病毒X蛋白结合蛋白(英简HBXIP)对肝癌细胞核因子(英简NF-KB)转录活性的影响。方法:应用基因共转染把NF-KB报告基因质粒HBXIP、pNF-KB-Luc真核表达载体(即为pcDNA3-hbxip)导入人肝癌细胞(H7402)系内,通过荧光素酶活性进行分析。结果:肝癌H7402细胞过表达HBXIP以后,明显提高了NF-KB转录活性;肝癌H7402细胞内HBXIP过表达以后,p65/NF-KB在细胞核内的含量显著增多。结论:乙肝病毒X蛋白结合蛋白能提高细胞核中p65/NK-KB蛋白的含量,具有良好的转录调控作用,以利于细胞增殖。
OBJECTIVE: To study the effect of hepatitis B virus X protein binding protein (HBV-B) on transcriptional activity of NF-κB in hepatocellular carcinoma cells. Methods: The eukaryotic expression vector of HBXIP and pNF-KB-Luc of NF-KB reporter gene (pcDNA3-hbxip) was introduced into human hepatoma cell line H7402 by gene co-transfection and analyzed by luciferase activity. Results: After HBXIP was overexpressed, the transcription activity of NF-κB in H7402 cells was significantly increased. After HBXIP was overexpressed in H7402 cells, the content of p65 / NF-κB in nucleus was significantly increased. Conclusion: HBV X protein-binding protein can increase the content of p65 / NK-KB protein in nucleus and has good transcriptional regulation to facilitate cell proliferation.
