目的:制备Heymann肾炎精细致病性抗原Gp330并复制人类膜性肾小球肾炎的动物模型。方法:用超速离心和钙沉降方法获取SD大鼠刷状缘微绒毛。用LCA-Sepharose4B亲合层析方法分离纯化Heymann抗原。用多点免疫方法获得抗体并免疫SD大鼠复制Heymann肾炎模型。结果:获取了Heymann肾炎的精细抗原Gp330并成功地用其抗体免疫SD大鼠,使其肾小球基底膜有免疫复合物沉积并产生大量蛋白尿。结论:Gp330是一种复制人类膜性肾小球肾炎模型的新抗原。
OBJECTIVE: To prepare a animal model of Heymann’s nephritis (PM) pathogenic Gp330 and replicate human membranous glomerulonephritis. Methods: Brush-like microvilli of SD rats were obtained by ultracentrifugation and calcium sedimentation. The Heymann antigen was isolated and purified by LCA-Sepharose 4B affinity chromatography. Antibody was obtained by multipoint immunization and immunized SD rats to replicate Heymann’s nephritis model. RESULTS: The G930, a fine antigen of Heymann’s nephritis, was obtained and successfully immunized SD rats with its antibody to deposit immune complex on the glomerular basement membrane and generate a large amount of proteinuria. Conclusion: Gp330 is a novel antigen that replicates the human membranous glomerulonephritis model.
