目的探讨通络保肾复方对肾小球系膜细胞(GMCs)表达转化生长因子-β(TGF-β)、纤维连接蛋白(FN)、层粘连蛋白(LN)的影响。方法通络保肾复方灌胃大鼠后,制备药物血清,将5%含药血清培养基加入体外培养的GMCs,脂多糖(LPS)作为刺激因子,将细胞分为空白组、LPS组、缬沙坦组及通络保肾复方小、中、大剂量组,刺激GMCs48h后,应用细胞免疫组化方法检测各组GMCs表达TGF-β、FN、LN的情况。结果通络保肾复方能够抑制GMCs表达TGF-β、FN、LN,其中以通络保肾复方大剂量组和中剂量组效果显著,与LPS组比较差异显著(P<0.05)。结论通络保肾复方可以抑制LPS诱导的GMCs表达TGF-β、FN、LN,从而延缓肾小球硬化的进程。
Objective To investigate the effect of Tonglu Baoshen Recipe on the expression of transforming growth factor-β (TGF-β), fibronectin (FN) and laminin (LN) in glomerular mesangial cells (GMCs). Methods The serum was prepared by intragastric administration of Tonglu Baoshen Compound, and 5% drug-containing serum was added into GMCs and lipopolysaccharide (LPS) as stimulating factors in vitro. The cells were divided into blank group, LPS group, After stimulating GMCs for48h, the expressions of TGF-β, FN and LN in GMCs in each group were detected by immunohistochemical method. Results Tongluo Baoshen Compound can inhibit the expression of TGF-β, FN and LN in GMCs. The effect of Tongluo Baoshen Compound high-dose group and middle-dose group is significant, which is significantly different from LPS group (P <0.05). Conclusion Tongluo Baoshen Recipe can inhibit the expression of TGF-β, FN and LN in LPS-induced GMCs and thus delay the progression of glomerulosclerosis.
