目的探讨染矽尘大鼠肺组织、肺泡灌洗液(BALF)和血清中基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)的表达变化。方法选取雄性Wistar大鼠84只随机分为实验组(100mg/ml)和生理盐水对照组,采用非气管暴露法建立大鼠矽肺模型,于染尘第3、7、14、21、28、60d处死大鼠,取肺组织并收集血清和BALF。免疫组化法检测肺组织中MMP-9和MMP-2的表达;酶联免疫吸附法(ELISA)检测血清和BALF中MMP-9和MMP-2的浓度。结果免疫组化染色结果,与对照组比较,MMP-9和MMP-2在染尘初期含量升高,且随着时间的延长呈增强趋势,分别在第14d和21d达到峰值;在染尘后期阳性表达仍较高,但与前几个时间点相比表达已减弱。ELISA结果显示,BALF和血清中MMP-9、MMP-2含量变化与肺组织中变化趋势相同,与染尘时间有相关关系(P<0.05),均呈先上升后下降的趋势,但血清和BALF中两指标相关性差(P>0.05)。结论 MMP-9、MMP-2参与了矽肺早期炎症及后期的纤维化过程,其表达随着时间的延长呈先上升后下降的趋势。
Objective To investigate the expression of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) in the lung tissue, BALF and serum of rats with silica dust. Methods Eighty-four male Wistar rats were randomly divided into experimental group (100mg / ml) and normal saline control group. Silicosis model was established by non-tracheal exposure. At the 3rd, 7th, 14th, 21st, Rats were sacrificed, lung tissue was taken and serum and BALF were collected. The expression of MMP-9 and MMP-2 in lung tissue was detected by immunohistochemistry. The concentrations of MMP-9 and MMP-2 in serum and BALF were detected by enzyme linked immunosorbent assay (ELISA). Results Compared with the control group, the content of MMP-9 and MMP-2 in the early stage of dust exposure increased with the increase of time and reached the peak on the 14th and 21th day respectively. The positive expression is still high, but the expression has weakened compared with previous time points. The results of ELISA showed that the changes of MMP-9 and MMP-2 in BALF and serum were the same as those in lung tissue, and were correlated with the dusting time (P <0.05) The correlation between the two indicators in BALF was poor (P> 0.05). Conclusion MMP-9 and MMP-2 are involved in the early stage of silicosis and fibrosis. The expression of MMP-9 and MMP-2 first increases and then decreases with the extension of time.
