目的:研究碳酸酐酶2(CA2)在高糖刺激下的表达改变和对心肌成纤维细胞凋亡、增殖的影响。方法:SD乳鼠心肌原代培养差速离心法分离心肌成纤维细胞,分为3组:正常糖(5.5 mmol/L,NG)组、高糖(25mmol/L,HG)组、高糖加碳酸酐酶抑制剂乙氧苯唑胺(ETZ)组。细胞免疫荧光观察CA2的表达变化,MTS测定心肌成纤维细胞的增殖,Western blot检测bax、bcl-2、caspase3、carbonic anhydrase2的蛋白表达情况。结果:与NG组相比,高糖能促进心肌成纤维细胞增殖和凋亡,并上调CA2的表达,通过抑制CA2可以抑制心肌成纤维细胞增殖和凋亡。结论:通过抑制CA2可减缓高糖诱导的心肌成纤维细胞增殖和凋亡。
Objective: To study the effect of carbonic anhydrase 2 (CA2) on the expression of cardiomyocyte stimulated by high glucose and on the apoptosis and proliferation of cardiac fibroblasts. Methods: Myocardial fibroblasts were isolated from SD rat by differential centrifugation and divided into 3 groups: normal glucose (5.5 mmol / L, NG), high glucose (25 mmol / L, HG) Carbonic anhydrase inhibitor ethoxazolam (ETZ) group. The expression of CA2 was observed by immunofluorescence, the proliferation of cardiac fibroblasts was measured by MTS, and the protein expression of bax, bcl-2, caspase3 and carbonic anhydrase2 was detected by Western blot. Results: Compared with NG group, high glucose could promote the proliferation and apoptosis of cardiac fibroblasts and up-regulate the expression of CA2. Inhibition of CA2 could inhibit the proliferation and apoptosis of cardiac fibroblasts. Conclusion: CA2 can inhibit the proliferation and apoptosis of cardiac fibroblasts induced by high glucose.
