碱提类肝素组分Ⅱ(简称C_2)是采用碱性工艺从猪十二指肠粘膜中提取的一种酸性粘多糖,对其抗血栓、调整血脂等方面已进行了大量的研究。本研究观察静脉注射C_2对动物脂蛋白脂酶(LPL)、肝脂酶(HL)及血浆脂质成分的影响。选用SD系大鼠、随机分为三组,分别给予生理盐水、肝素和C_2(给药剂量两者折合抗凝效价基本相同,为250u/kg)。用药前及药后15分钟分别心脏取血,分离血浆测定LPL及HL活性。结果发现静脉注射C_2可使血浆LPL及HL活性升高,且C_2作用强于肝素。另选用健康雄性家兔,分组及给药剂量同前,同法取血制备血浆,分别测定LPL、HL活性。酶法测定血浆中TC、HDL-ch,计算(VLDL+LDL)-ch和HDL-ch/TC。结果表明,C2可升高血浆中两种脂酶活性,HDL-ch和HDL-ch/TC,降低(VLDL+LDL)-ch和TC,HDL-ch升高和(VLDL+LDL)-ch降低与LPL活性呈相关,提示用药后血浆脂质变化由药物释放脂酶所致。
Alkalin Heparin Component Ⅱ (C_2) is an acidic mucopolysaccharide extracted from porcine duodenal mucosa by alkaline process. A large amount of studies have been conducted on its antithrombotic and lipid-lowering effects. This study was to observe the effects of intravenous C_2 on lipoprotein lipase (LPL), hepatic lipase (HL) and lipid composition in plasma. SD rats were randomly divided into three groups and given normal saline, heparin and C_2 respectively (the dosage of the two was equivalent to the same anticoagulant titer of 250u / kg). Blood samples were taken from the heart before and 15 minutes after drug administration, respectively, and plasma levels of LPL and HL were measured. The results showed that intravenous C_2 plasma LPL and HL activity increased, and C_2 stronger than heparin. Another selection of healthy male rabbits, grouping and administration of the same dose, with the same method to prepare blood plasma, were measured LPL, HL activity. Enzymatic determination of plasma TC, HDL-ch, calculated (VLDL LDL) -ch and HDL-ch / TC. The results showed that C2 increased both lipase activity, HDL-ch and HDL-ch / TC, decreased (VLDL + LDL) -ch and TC, increased HDL-ch and decreased (VLDL + LDL) And LPL activity was correlated, suggesting that changes in plasma lipids after drug release of lipase.
