目的探讨四氧化三铁纳米(nano-Fe_3O_4)颗粒对大鼠肠道内短链脂肪酸影响。方法 60只(雌雄各半)SD大鼠随机分为3组,分别为5%纤维素钠对照组、低、高剂量nano-Fe_3O_4组,每组20只,雌雄各半,每日灌胃给予不同剂量nano-Fe_3O_4,连续4周,每周以逼迫法采集新鲜粪便样品,采用气相色谱法测定大鼠肠道内的短链脂肪酸含量。结果染毒第4周时,与对照组比较,低、高剂量nano-Fe_3O_4组雌性大鼠肠道内丁酸含量[分别为(0.05±0.02)、(0.06±0.03)μg/μL]明显下降(P<0.05),低、高剂量nano-Fe_3O_4组雄性大鼠肠道内乙酸含量[分别为(1.31±0.16)、(1.54±0.22)μg/μL]明显下降(P<0.05),高剂量nano-Fe_3O_4组雄性大鼠肠道内丁酸含量[(0.04±0.02)μg/μL]下降,差异有统计学意义(P<0.05);染毒组大鼠部分肠上皮细胞出现空泡变性,杯状细胞排列疏松,柱状吸收细胞减少。结论此实验条件下nano-Fe_3O_4颗粒可能对大鼠肠道内短链脂肪酸含量产生影响。
Objective To investigate the effect of nano-Fe 3 O 4 nanoparticles on short-chain fatty acids in intestines of rats. Methods Sixty male and female SD rats were randomly divided into 3 groups: 5% sodium cellulose control group, low and high dose nano-Fe 3 O 4 groups, 20 rats in each group, Different doses of nano-Fe 3 O 4, for 4 consecutive weeks, were forced to law every week to collect fresh fecal samples, gas chromatography was used to determine the content of short-chain fatty acids in the intestine of rats. Results Compared with the control group, the content of butyric acid in the intestinal mucosa of the rats in low and high dose groups of nano-Fe 3 O 4 [(0.05 ± 0.02) and (0.06 ± 0.03) μg / μL] was significantly decreased at the 4th week (P <0.05). The contents of acetic acid in the intestinal tract of low and high dose nano-Fe 3 O 4 groups were significantly decreased (1.31 ± 0.16 and 1.54 ± 0.22 μg / μL, respectively, P <0.05) The content of butyric acid in the intestinal tract of Fe_3O_4 group decreased ([(0.04 ± 0.02) μg / μL], the difference was statistically significant (P <0.05); some intestinal epithelial cells were vacuolar degeneration, Lined loose, columnar absorption of cells decreased. Conclusion Under the experimental conditions, nano-Fe 3 O 4 particles may affect the short-chain fatty acids in the intestine of rats.
