目的建立灭菌定性试验中和剂鉴定方法。方法按照评价灭菌效果的试验体系和试验过程,直接在中和剂肉汤、中和产物肉汤中加入试验菌的方法进行实验研究。结果含500 mg/L硫代硫酸钠肉汤及其与≤3 000 mg/L过氧乙酸作用后形成的中和产物肉汤对枯草杆菌黑色变种芽孢的恢复生长无影响。含2 000 mg/L硫代硫酸钠肉汤及其与≤5 000 mg/L过氧乙酸作用后形成的中和产物肉汤可恢复大量枯草杆菌黑色变种芽孢的生长,但对少量的枯草杆菌黑色变种芽孢的恢复生长有影响。含≥3 000 mg/L硫代硫酸钠肉汤与任何浓度的过氧乙酸作用形成的中和产物肉汤对枯草杆菌黑色变种芽孢的恢复生长均有影响。结论按照评价灭菌定性效果的试验体系和试验过程建立的灭菌定性中和剂鉴定方法,是一种科学、合理、准确的中和剂鉴定方法。灭菌定性试验中,应严格鉴定中和剂的有效性和对试验菌恢复生长的影响。
OBJECTIVE To establish a method for the neutralization of qualitative test of sterilization. Methods According to the test system and test process of evaluating the sterilization effect, the experimental bacteria were directly added into the broth of neutralizer and the broth of neutralization. The results showed that 500 mg / L sodium thiosulfate broth and its neutralization product broth formed after the effect of ≤3000 mg / L peracetic acid had no effect on the growth recovery of B. subtilis black spores. Broth containing 2,000 mg / L sodium thiosulfate and its neutralization product broth formed after the effect of ≤5,000 mg / L peracetic acid could restore a large number of B. subtilis black spores, but a small amount of Bacillus subtilis Black variant spores have an impact on the restoration and growth. The broth of neutralization product with ≥3000 mg / L sodium thiosulfate broth and any concentration of peracetic acid formed influence on the recovery and growth of B.subtilis black variant spores. Conclusion According to the experimental system for evaluating the qualitative effect of sterilization and the identification method of sterilized neutralizing agent established during the experiment, it is a scientific, reasonable and accurate method for the identification of neutralizer. Sterilization qualitative test, should be strictly identified the effectiveness of neutralizers and test the impact of restoration of growth.
