目的:研究中药菝葜中的降血糖活性成分及对α-葡萄糖苷酶和醛糖还原酶的抑制作用。方法:采用有机溶剂萃取、硅胶柱层析等分离纯化技术进行降血糖活性成分的分离纯化,并通过理化性质和波谱数据鉴定活性物质的结构,继而通过动物实验模型和酶-抑制剂模型研究该活性物质的降血糖作用和酶学性质。结果:从菝葜中分离并鉴定出一种具有显著降血糖活性的化合物,结构为3(S)-5,7,4’-三羟基二氢黄酮(柚皮素),具有α-葡萄糖苷酶和醛糖还原酶抑制活性。结论:柚皮素为首次从菝葜中分离得到的降血糖有效成分,其显著的酶抑制活性为进一步阐明菝葜的降血糖机制提供了参考。
OBJECTIVE: To study the hypoglycemic active ingredients of Radix Angelicae Dahuricae and its inhibitory effects on α-glucosidase and aldose reductase. METHODS: The separation and purification of active components for hypoglycemic use were carried out by organic solvent extraction and silica gel column chromatography. The structures of the active substances were identified by physicochemical properties and spectral data. The animal model and enzyme-inhibitor model were then used to study the structure of the active substances. The hypoglycemic and enzymatic properties of the active substance. RESULTS: A compound with significant hypoglycemic activity was isolated and identified from Radix Astragali with the structure 3(S)-5,7,4’-trihydroxydihydroflavone (naringenin) with alpha-glucoside. Enzymes and aldose reductase inhibitory activity. Conclusion: Naringenin is the effective component of hypoglycemia isolated from quail for the first time. Its significant enzyme inhibitory activity provides a reference for further elucidating the hypoglycemic mechanism of alfalfa.
