目的探讨诱导性多能干细胞对造血衰竭小鼠模型进行造血重建的作用。方法建立雌性小鼠造血衰竭模型,移植经天然活性物质诱导的雄性小鼠脾细胞,检测受体小鼠生存时间、外周血WBC、脾湿重及脾集落(CFU-S)数,进行G带染色体分析受体小鼠是否有Y染色体。结果经天然活性物质诱导的雄性小鼠脾细胞5×106个输注给致死剂量照射后的雌性小鼠,可明显延长其存活时间,提高受体小鼠外周血WBC数,也能增加脾湿重,提高脾集落数,经G带染色体分析进行Y染色体检测,受体小鼠骨髓检测可观察到20%的Y染色体。结论经天然活性物质诱导的小鼠脾细胞含有多能干细胞,回输注入造血衰竭的小鼠能重建小鼠造血功能。
Objective To investigate the role of induced pluripotent stem cells in hematopoietic reconstitution in hematopoietic failure mouse model. Methods The model of hematopoietic failure in female mice was established. Splenocytes of male mice induced by natural active substances were transplanted. The survival time of recipient mice, WBC in peripheral blood, wet weight of spleen and number of spleen colonies (CFU-S) Chromosome analysis of recipient mice for Y chromosome. Results The 5 × 106 splenocytes of male mice induced by natural active substances could significantly prolong the survival time and increase the number of WBC in peripheral blood of recipient mice, Heavy, increase the number of spleen colonies, the G chromosome chromosome analysis of Y chromosome detection, bone marrow detection of recipient mice observed 20% Y chromosome. Conclusion The mouse spleen cells induced by natural active substances contain pluripotent stem cells, and the mice injected with hematopoietic failure can reconstruct the hematopoietic function in mice.
