目的 :探讨一氧化氮 (nitric oxide NO)对肝硬化肝脏缺血再灌注 (ischemia- reperfu-sion I/ R)损伤后肝细胞 P-选择素表达的影响及保护作用。方法 :建立肝硬化大鼠肝脏缺血再灌注模型 ,观察 L -精氨酸 (L - Arginine)、Nω-硝基 - L -精氨酸甲酯 (L - NAME)预处理组和 I/ R组肝细胞 P-选择素表达情况以及肝功能损伤情况。结果 :I/ R对肝细胞 P-选择素表达增强 (P<0 .0 1) ,肝损伤加重。NO供体 - L - Arginine预处理后 P-选择素表达下降 (P<0 .0 1) ,并在一定程度上减轻肝 I/ R损伤程度 ,而一氧化氮合酶抑制剂 - L - NAME预处理后 P-选择素表达增强明显 (P<0 .0 1) ,对肝脏无保护作用。结论 :肝硬化肝脏 I/ R损伤与肝细胞 P-选择素的表达有关 ,NO能抑制肝细胞 P-选择素的表达 ,减轻肝脏 I/ R损伤程度
Objective: To investigate the effect and the protective effect of nitric oxide (NO) on the expression of P-selectin in hepatocytes after liver ischemia-reperfu-sion I / R injury in cirrhotic rats. Methods: The model of hepatic ischemia - reperfusion in cirrhotic rats was established. The effects of L - arginine, Nω - nitro - L - arginine methyl ester (L - NAME) Group P-selectin expression of liver cells and liver damage. Results: I / R enhanced the expression of P-selectin in hepatocytes (P <0.01) and aggravated hepatic injury. The expression of P - selectin decreased after NO donor - L - Arginine pretreatment (P <0.01), and reduced the degree of hepatic I / R injury to a certain degree. However, the nitric oxide synthase inhibitor L - NAME P-selectin expression increased significantly after pretreatment (P <0.01), no protective effect on the liver. CONCLUSIONS: Liver I / R injury in cirrhotic patients is related to the expression of P-selectin in hepatocytes. NO can inhibit the expression of P-selectin in hepatocytes and reduce the degree of hepatic I / R injury
