目的观察山茱萸多糖对亚急性衰老大鼠睾丸形态及超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)的影响。方法 2014年3—6月开展实验,取清洁级雄性SD大鼠40只,随机数字表法分为对照组、模型组、山茱萸多糖组、雄激素组,每组10只。采用D-半乳糖200 mg/(kg·d)腹腔注射6周构建大鼠亚急性衰老模型,同时对照组、模型组每天给予生理盐水,山茱萸多糖组、雄激素组分别给予山茱萸多糖50 mg/(kg·d)、雄激素5 mg/(kg·d)灌胃6周。紫外分光光度计法检测睾丸组织中SOD、MDA的含量。HE染色观察睾丸形态结构。计数资料比较用方差分析,组间两两比较用LSD-t检验,P<0.05为差异有统计学意义。结果与对照组SOD活性、MDA水平[(119.43±11.62)U/mgprot、(0.69±0.19)nmol/mgprot]相比,衰老大鼠睾丸组织SOD活性[(104.67±09.85)U/mgprot]明显下降,MDA水平[(1.39±0.59)nmol/mgprot]显著升高,对比差异均有统计学意义(均P<0.05),山茱萸多糖组、雄激素组睾丸组织SOD活性[(116.74±10.06)、(117.38±10.89)U/mgprot]明显升高,MDA水平[(0.89±0.39)、(0.85±0.38)nmol/mgprot]显著降低,对比差异均有统计学意义(均P<0.05)。结论山茱萸多糖对衰老大鼠睾丸具有改善作用,其机制可能与增加睾丸组织SOD的活性,减少MDA的含量有关。
Objective To observe the effects of Fructus Corni polysaccharide on testicular morphology and superoxide dismutase (SOD), malondialdehyde (MDA) in subacute aging rats. Methods From March to June in 2014, 40 clean male SD rats were randomly divided into control group, model group, dogwood polysaccharide group and androgen group, with 10 rats in each group. The model of subacute aging was established by intraperitoneal injection of D-galactose 200 mg / (kg · d) for 6 weeks. At the same time, the rats in the control group and the model group were treated with saline, Fructus Corni polysaccharide group, and Fructus corni polysaccharide 50 mg / (kg · d) and androgen 5 mg / (kg · d) for 6 weeks. Counting data were compared using ANOVA, LSD-t test was used to compare the two groups, P <0.05 was considered statistically significant. Results Compared with the control group, the SOD activity in testis tissue [(104.67 ± 09.85) U / mgprot] was significantly decreased compared with that in control group [(119.43 ± 11.62) U / mgprot, (0.69 ± 0.19) nmol / , MDA level (1.39 ± 0.59) nmol / mgprot] were significantly increased, the differences were statistically significant (all P <0.05), SOD activity in the testis tissue of Fructus Corni polysaccharide group and androgen group [(116.74 ± 10.06), 117.38 ± 10.89) U / mgprot] were significantly higher than those in control group (P <0.05). The level of MDA was significantly lower than that of control group (0.89 ± 0.39 vs 0.85 ± 0.38 nmol / mgprot). Conclusion Fructus Corni polysaccharide can improve the testis of aging rats. The mechanism may be related to increasing the activity of SOD and decreasing the content of MDA in testis.
