以蓝莓产区发现的一种新病害蓝莓叶穿孔病样为供试病菌,采用柯赫氏证病法则、形态学测定、ITS和EF-1α序列比对鉴定和常规病原菌生物学特性研究技术,研究了该病的致病菌,致病菌的分类地位、致病菌在不同培养条件下菌丝生长和孢子萌发的特性。结果表明:引起蓝莓叶穿孔病的致病菌为Alternaria alternata,该菌在供试条件下菌落生长以PDA为最优培养基,30℃为最佳生长温度,最佳pH 5,最佳氮源为甘氨酸,最佳碳源为葡萄糖;孢子萌发最佳条件为30℃,pH 5,2%甘氨酸,2%葡萄糖。病原菌菌丝致死温度为56℃、10min。
A new disease was found in the blueberry producing area. Blueberry leaf perforation was used as the pathogen for pathogenesis. Koch’s disease rule, morphology, ITS and EF-1α sequence alignment and routine pathogenic bacteria biological characteristics were studied. The pathogens of the disease, the classification of pathogenic bacteria, the growth of mycelium and spore germination of pathogenic bacteria under different culture conditions were studied. The results showed that the pathogen causing blueberry leaf perforation was Alternaria alternata. Under the conditions of the test, the colony growth of the strain was optimized by PDA. The best growth temperature was 30 ℃, the optimum pH was 5, the best nitrogen source As the glycine, the best carbon source was glucose. The optimum conditions for spore germination were 30 ℃, pH 5, 2% glycine and 2% glucose. Pathogenic mycelial lethal temperature 56 ℃, 10min.
