目的探讨游离移植后长期存活的兔颌下腺细胞的生物学特性。设计实验性研究。研究对象兔颌下腺细胞。方法将移植成功后存活8个月的兔取出移植的颌下腺,进行细胞培养,观察其细胞生长特性,进行流式细胞仪分析及甲基噻唑基四唑(MTT)检测,并与培养的正常颌下腺细胞进行对比。主要指标细胞生长特性、流式细胞仪分析、细胞生长曲线。结果移植颌下腺细胞组与正常颌下腺细胞组具有相似的细胞形态和生长特性,移植颌下腺细胞组细胞周期的G1期、S期及G2期各时相细胞增值百分比分别是(82.7±2.10)%、(6.2±0.98)%、(11.2±1.24)%,正常颌下腺细胞组各时相分别是(82.0±2.06)%、(6.3±1.12)%、(11.7±1.35)%,两组相比较无明显差异(P均>0.05),同时两组具有相似的细胞生长曲线。结论游离移植后长期成活的兔颌下腺细胞与非移植的正常颌下腺细胞具有相近的细胞生长特性,去除神经支配后游离移植的颌下腺细胞的生物学特性未发生明显改变。
Objective To investigate the biological characteristics of long-term survival of rabbit submandibular gland cells after free transplantation. Design experimental research. Rabbit submandibular gland cells Methods The transplanted submandibular glands were removed after transplantation for 8 months and the cell growth was observed. The growth characteristics of the submandibular glands were observed. The growth of the submandibular gland was observed by flow cytometry and methyl thiazolyl tetrazolium (MTT) assay. Cells were compared. The main indicators of cell growth characteristics, flow cytometry analysis, cell growth curve. Results The submandibular gland cells had similar cell morphology and growth characteristics with those of normal submandibular gland cells. The cell proliferation rates in G1 phase, S phase and G2 phase of the submandibular gland cells were (82.7 ± 2.10)%, ( (11.2 ± 1.98)%, (11.2 ± 1.24)%, respectively. The time of normal submandibular gland cell group was (82.0 ± 2.06)%, (6.3 ± 1.12)%, (11.7 ± 1.35)% respectively. There was no significant difference between the two groups (P> 0.05), while the two groups had similar cell growth curves. CONCLUSION: Long-term survival of rabbit submandibular gland cells and normal submandibular gland cells after free transplantation have similar cell growth characteristics. The biological characteristics of free submandibular gland cells after transplantation have no significant changes.
