研究以霍乱毒素B亚基 (CTB)为载体的重组疟疾多价抗原 (AWTE)表位的DNA疫苗在恒河猴中的免疫原性及对相应疟原虫感染的免疫保护作用。方法 :以恒河猴为模型 ,肌肉途径免疫编码CTB AWTE基因的质粒 ,免疫间隔为 15d ,免疫 3次 ,免疫剂量为每种质粒每次 50 0 μg/只。结果 :DNA疫苗组免疫 2次后即产生了较高水平的细胞免疫和体液免疫 ,免疫后 91d用 1.2 5× 10 8个猴疟原虫攻击 ,对照组 5只动物在攻击后 14d左右全部感染 ,感染持续 34d以上 ;DNA疫苗组的 5只动物在攻击后 60d没有感染。结论 :这种鸡尾酒式的抗原表位组合构建的DNA疫苗具有很好的免疫原性 ,同时也说明DNA疫苗在抗疟感染中起着举足轻重的作用。
To study the immunogenicity and immunoprotective effect of DNA vaccine of recombinant malaria polyvalent antigen (AWTE) epitope using Cholera toxin B subunit (CTB) as carrier in rhesus monkeys. Methods: The model of rhesus macaque was used to immunize the muscle of CTB AWTE gene. The immune interval was 15 days, immunized three times, and the immunized dose was 50 μg / mouse for each plasmid. Results: After immunized with DNA vaccine for two times, high levels of cellular immunity and humoral immunity were produced. After immunization, 1.2 5 × 10 8 P. falciparum was challenged 91 days after immunization, and all animals in control group were infected at 14 days after challenge. The infection lasted for more than 34 days. Five animals in the DNA vaccine group were not infected 60 days after the challenge. Conclusion: The DNA vaccine constructed by combining this cocktail epitope has good immunogenicity and at the same time shows that DNA vaccine plays an important role in anti-malarial infection.
