目的:建立疏花石斛多糖提取优选方案。方法:采用传统水浸提法、纤维素酶法、超声辅助法和微波辅助法4种不同处理方法对疏花石斛多糖进行提取;多糖含量的测定用苯酚-硫酸法。结果:纤维素酶法较优方案为1∶30料液比,50℃,pH 4.5,200 U·mL-1作用2 h;微波辅助法较优方案为料液比1∶30、微波功率档位为中火、作用时间8 min预处理后,80℃浸提1.5 h;超声辅助法较优方案为料液比1∶30、超声功率300 W、超声处理6 min预处理后,80℃浸提1.5 h;传统水浸提法较优方案为料液比1∶30、温度80℃浸提1.5 h。结论:纤维素酶法提取疏花石斛多糖效果最佳,是多糖提取时的首选方案,传统水浸提法宜结合超声辅助法和微波辅助法。
Objective: To establish the optimal solution of polysaccharides from Dendrobium candidum. Methods: The polysaccharides of Dendrobium officinale were extracted by four different methods: traditional water extraction, cellulase, ultrasonic assisted and microwave assisted method. The content of polysaccharides was determined by phenol-sulfuric acid method. Results: The optimum conditions of cellulase were as follows: 1:30 ratio of solid to liquid, 50 ℃, pH 4.5, 200 U · mL-1 for 2 h. The optimum microwave assisted method was the ratio of solid to liquid 1:30, microwave power After the pretreatment at 8 min for 8 min and the extraction at 80 ℃ for 1.5 h, the optimal solution of ultrasonic assisted method was the pretreatment at the material-liquid ratio of 1:30, the ultrasonic power of 300 W and the ultrasonic treatment of 6 min, and the pretreatment at 80 ℃ Mention 1.5 h; the traditional water-extraction method for the optimal solution ratio of 1:30, extraction temperature of 80 ℃ 1.5 h. CONCLUSION: The optimal extraction of polysaccharides from Dendrobium candidum by cellulase is the first choice for polysaccharide extraction. The traditional water-extraction method should be combined with ultrasound-assisted method and microwave-assisted method.
