目的 研究润坦对局灶性脑缺血再灌注的神经保护效应。方法 制备线栓法大脑中动脉闭塞再灌注模型,干预组每天给予润坦3mg/kg,对照组给予等量生理盐水,每日对大鼠神经功能缺损进行评分,于再灌注12h、24h、48h和5d处死动物,切片作TUNEL原位凋亡检测。结果 润坦可以显著改善术后第2d大鼠的神经功能缺损(P<0.05),降低致死率,减少再灌注24h以后的细胞凋亡数目(P<0.05)。结论 润坦可以抑制大鼠脑缺血再灌注后神经元凋亡,对急性期脑梗死可能有神经保护效应。
Objective To study the neuroprotective effects of Runtan on focal cerebral ischemia and reperfusion. Methods The model of occlusion of middle cerebral artery occlusion was prepared by thread embolism. The intervention group was treated with Runtan 3mg/kg every day. The control group was given the same amount of normal saline and the neurological deficit was scored daily. The reperfusion was performed at 12h, 24h and 48h. The animals were sacrificed at 5 days and sectioned for TUNEL in situ apoptosis detection. Results Runtan significantly improved the neurological deficits (P<0.05), decreased the lethality, and reduced the number of apoptotic cells after 24 hours of reperfusion (P<0.05). Conclusion Runtan can inhibit neuronal apoptosis after cerebral ischemia-reperfusion in rats, and may have neuroprotective effect on acute cerebral infarction.
