目的:探讨TLR9(Toll-like receptor 9)在小鼠结直肠癌(Colorectal cancer,CRC)形成早期过程中及在临床CRC样品中的表达水平。方法:通过予以C57BL/6小鼠腹腔注射氧化偶氮甲烷(Azoxymethane,AOM)后接以3轮的葡聚糖硫酸钠(Dextran sulfate sodium,DSS)处理建立炎症相关性结直肠癌模型,采集肿瘤形成早期(第14天)结直肠组织,通过q RT-PCR方法检测TLR9基因在小鼠结肠中的表达。另外,通过Western-blot和q RT-PCR检测26对临床新鲜结直肠癌组织及相应癌旁组织TLR9的表达水平。结果:与对照组相比,TLR9在小鼠结直肠癌形成早期及人结直肠癌组织中的表达均呈显著性下降(P﹤0.05)。结论:结直肠中TLR9低表达可能是CRC的促因之一,TLR9可能具有抑制CRC的发生发展和治疗CRC的潜能。
Objective: To investigate the expression of TLR9 (Toll-like receptor 9) in the early stage of colorectal cancer (CRC) formation and in clinical CRC samples. Methods: C57BL / 6 mice were injected intraperitoneally with Azoxymethane (AOM) and then treated with Dextran sulfate sodium (DSS) for 3 days to establish an inflammation-associated colorectal cancer model. The tumors were collected Early (day 14) colorectal tissue was formed and the TLR9 gene expression in the mouse colon was examined by q RT-PCR. In addition, the expression of TLR9 in 26 clinical fresh colorectal cancer tissues and corresponding paracancerous tissues was detected by Western-blot and q RT-PCR. Results: Compared with the control group, the expression of TLR9 in the early stage of colorectal cancer and colorectal cancer was significantly decreased (P <0.05). Conclusion: The low expression of TLR9 in colorectal may be one of the promoters of CRC. TLR9 may have the potential of inhibiting the development of CRC and treating CRC.
