目的探讨中药四物消风颗粒对变应性接触性皮炎(allergic contact dermatitis,ACD)小鼠免疫器官胸腺、脾脏及其T、B淋巴细胞增殖的影响。方法将60只小鼠完全随机分为6组,分别为正常对照组,ACD模型对照组,氢化可的松阳性对照组及四物消风颗粒高、中、低剂量组,每组10只。观测各组小鼠耳肿胀程度、胸腺指数、脾脏指数以及丝裂原诱导的脾T、B淋巴细胞的增殖反应。结果与模型对照组小鼠耳肿胀程度比较,氢化可的松组,四物消风颗粒低、中、高剂量组小鼠耳肿胀程度差异均有统计学意义(P<0.05)。两两比较显示四物消风颗粒中、高剂量组优于低剂量组(P<0.05),与阳性对照组比较无明显差异(P>0.05)。与模型对照组小鼠胸腺指数、脾脏指数比较,氢化可的松阳性对照组、四物消风颗粒低剂量组、中剂量组、高剂量组均显著减低(P<0.05),四物消风颗粒高、中剂量组优于低剂量组(P<0.05),四物消风颗粒高剂量组与阳性对照组无明显差异(P>0.05)。与模型对照组脾脏T、B淋巴细胞增殖活性比较,氢化可的松阳性对照组、四物消风颗粒低剂量组、中剂量组、高剂量组差异均有统计学意义(P<0.05)。同时两两比较显示四物消风颗粒中、高剂量组优于低剂量组(P<0.05),与阳性对照组比较无明显差异(P>0.05)。结论四物消风颗粒防治小鼠ACD的药理机制可能与其调节T、B淋巴细胞功能作用有关。
Objective To investigate the effect of Siwu Xiaofeng Granule on the proliferation of thymus and spleen and T and B lymphocytes of immune organs in mice with allergic contact dermatitis (ACD). Methods Sixty mice were randomly divided into six groups, namely normal control group, ACD model control group, hydrocortisone positive control group and Siwu Xiaofeng Granules in high, medium and low dose groups, 10 in each group. The degree of ear swelling, thymus index, spleen index and mitogen-induced proliferation of splenic T and B lymphocytes in each group were observed. Results Compared with mice in the model control group, the degree of ear edema in mice treated with hydrocortisone and Siwu Xiaofeng Granules was significantly different (P <0.05). The comparison of two groups showed that Siwu Xiaofeng Granules was higher than the low-dose group (P <0.05), and no significant difference compared with the positive control group (P> 0.05). Compared with the model control group, the thymus index and spleen index of the mice in the control group were significantly decreased (P <0.05), the hydrocortisone positive control group, the Siwu Xiaofeng Granule low dose group, the middle dose group and the high dose group were significantly decreased (P <0.05) There was no significant difference between the high dosage of Siyu Xiaofeng Granules and the positive control group (P> 0.05). Compared with model control group, the proliferation activity of T and B lymphocytes in spleen was significantly lower in hydrocortisone positive control group, Siwu Xiaofeng Granules low dose group, middle dose group and high dose group (P <0.05). Simultaneous comparison of two groups showed that Siwu Xiaofeng Granules was higher than the low dose group (P <0.05), and no significant difference compared with the positive control group (P> 0.05). Conclusion The pharmacological mechanism of Siwu Xiaofeng Granule in preventing and treating ACD in mice may be related to regulating the function of T and B lymphocytes.
