目的探讨二烯丙基三硫(DATS)对急性肺损伤小鼠血清IL-18,IL-10的影响。方法 90只健康昆明小鼠,随机分为5组:①急性肺损伤组:腹腔注射大肠杆菌脂多糖(LPS);②预防组:腹腔注射DATS 7d后注射LPS;③治疗组:腹腔注射LPS后30min注射DATS;④DATS组:腹腔注射DATS 7d;⑤对照组:腹腔注射等量生理盐水。光镜观察12h点肺组织形态学改变。测定血清中IL-18及IL-10在2h和6h含量。结果 DATS预防组肺泡间渗出及炎性细胞浸润较ALI组明显减轻。其血清中IL-18含量均明显低于ALI组(P<0.01),IL-10含量均明显高于ALI组(P<0.01)。结论预防性应用DATS可减轻LPS诱导的ALI小鼠肺组织炎症反应。并能抑制LPS诱导的小鼠血清中炎性细胞因子IL-18的生成,而促进抗炎性细胞因子IL-10的生成。
Objective To investigate the effect of diallyl trisulfide (DATS) on serum IL-18 and IL-10 in mice with acute lung injury. Methods 90 healthy Kunming mice were randomly divided into 5 groups: ① acute lung injury group: intraperitoneal injection of E.coli lipopolysaccharide (LPS); ② prophylaxis group: intraperitoneal injection of DATS 7d after injection of LPS; ③ treatment group: after intraperitoneal injection of LPS 30 min injection of DATS; ④ DATS group: intraperitoneal injection of DATS 7d; ⑤ control group: intraperitoneal injection of normal saline. Light microscopy 12h point lung morphology changes. Serum IL-18 and IL-10 levels at 2h and 6h were determined. Results In the DATS prevention group, alveolar exudate and inflammatory cell infiltration were significantly reduced compared with those in the ALI group. The content of IL-18 in serum was significantly lower than that in ALI group (P <0.01), IL-10 content was significantly higher than that in ALI group (P <0.01). Conclusion The prophylactic application of DATS can attenuate the lung inflammation induced by LPS in ALI mice. And can inhibit LPS-induced mouse sera inflammatory cytokine IL-18 production, and promote the anti-inflammatory cytokine IL-10 production.
