目的了解新基因AI408225与大鼠肝再生涉及的体液免疫相关性。方法大鼠再生肝8种细胞的分离,采用Percoll密度梯度离心结合免疫磁珠分选方法进行,用MicrosoftExcel、BLAST等软件分析基因的序列同源性及共表达关系,用生物信息学和系统生物学等方法分析新基因参与的生理活动。结果93个参与体液免疫的已知基因与肝再生相关,肝细胞、胆管上皮细胞、卵圆细胞、肝星形细胞、窦内皮细胞、库普弗细胞、陷窝细胞、树突状细胞的体液免疫相关基因数为33、46、17、59、48、35、53、68。其中,cd4与AI408225同源和共表达。结论大鼠肝再生与体液免疫相关,AI408225参与MHCII-抗原肽-CD4-TCR复合物形成。
Objective To investigate the correlation between humoral immunity and the new gene AI408225 involved in liver regeneration in rats. METHODS: Eight kinds of cells were isolated from rat regenerative liver using Percoll density gradient centrifugation combined with magnetic bead sorting. The sequence homology and coexpression were analyzed by software such as Microsoft Excel and BLAST. Among them, cd4 and AI408225 homologous and co-expression. Conclusion Rat liver regeneration is associated with humoral immunity. AI408225 is involved in the formation of MHC II-antigen peptide-CD4-TCR complex.
