目的研究Bcl-2结合抗凋亡基因(Bag-1)和鞘氨醇激酶-1(Spk-1)在胃癌组织中的表达及作用。方法 128例胃癌患者,用免疫组化的方法检测Bag-1、Spk-1蛋白在胃癌组织中的表达,探讨两种基因的表达与胃癌临床病理之间的关系。结果 Bag-1、Spk-1在胃癌细胞的胞浆和胞核中均有不同程度的表达,其中前者阳性表达较强,后者相对较弱。两种基因的表达与胃癌的分化程度和淋巴转移相关。结论 Bag-1及Spk-1上调可以抑制胃癌细胞凋亡,促进胃癌的分化和淋巴转移,两种基因具有协同作用。
Objective To study the expression and effect of Bcl-2 binding anti-apoptosis gene (Bag-1) and sphingosine kinase-1 (Spk-1) in gastric cancer. Methods The expression of Bag-1 and Spk-1 protein in gastric cancer tissues was detected by immunohistochemical method in 128 patients with gastric cancer. The relationship between the expression of two genes and clinicopathology of gastric cancer was explored. RESULTS: Bag-1 and Spk-1 were expressed in different degrees in the cytoplasm and nucleus of gastric cancer cells. The positive expression of Bag-1 and Spk-1 was relatively strong, and the latter was relatively weak. The expression of the two genes is related to the degree of differentiation and lymphatic metastasis of gastric cancer. Conclusion The upregulation of Bag-1 and Spk-1 can inhibit the apoptosis of gastric cancer cells and promote the differentiation and lymphatic metastasis of gastric cancer. The two genes have synergistic effects.
