目的分析子宫内膜癌I型和II型全基因表达谱。方法利用6张Affymetrix全基因组基因芯片,分析两者之间在表达谱上的差异,结合生物信息分析。结果许多基因明显在雌激素依赖型子宫内膜癌中高表达,其中,一些基因涉及了雌激素依赖型中雌激素的代谢与转化,另外一些基因可能为雌激素调节基因。CYP2C9参与了雌激素酮硫酸盐转化为16-羟基硫酸盐代谢;DDC能与类固醇受体结合,增强类固醇受体的转录活性。结论某些基因在雌激素依赖型子宫内膜癌中高表达,为进一步研究此类基因在子宫内膜癌发病过程的作用及预后奠定了基础。
Objective To analyze endometrial cancer type I and type II whole gene expression profiles. Methods Six Affymetrix genome-wide cDNA microarrays were used to analyze the difference in expression profile between the two, combined with bioinformatics analysis. Results A number of genes were clearly expressed in estrogen-dependent endometrial carcinomas. Some of these genes are involved in the metabolism and transformation of estrogen in estrogen-dependent type. Others may be estrogen-regulated genes. CYP2C9 is involved in the conversion of estrone sulfate to 16-hydroxy sulfate metabolism; DDC binds to steroid receptors and enhances the transcriptional activity of steroid receptors. Conclusion Some genes are highly expressed in estrogen-dependent endometrial carcinoma, which lays the foundation for further study on the role and prognosis of these genes in the pathogenesis of endometrial carcinoma.
