目的:对8个染料木黄酮衍生物抗血管内皮氧化应激损伤的活性进行筛选。方法:采用全自动生化分析仪测定8个染料木黄酮衍生物对氧化应激损伤的血管内皮细胞乳酸脱氢酶释放的影响,PI染色FCM检测8个染料木黄酮衍生物对内皮细胞凋亡的影响。结果:在所有衍生物中,7-二氟亚甲基-5,4-二甲氧基染料木黄酮抗血管内皮氧化应激损伤的活性最高,且其作用强于先导化合物染料木黄酮。结论:通过筛选获得的7-二氟亚甲基-5,4-二甲氧基染料木黄酮具有更强的抗血管内皮氧化应激损伤作用,具有进一步研究的价值。
OBJECTIVE: To screen the anti-oxidative stress effects of 8 genistein derivatives on vascular endothelial cells. Methods: The effect of 8 genistein derivatives on the release of lactate dehydrogenase (LDH) in endothelial cells injured by oxidative stress was determined by automatic biochemical analyzer. The effect of 8 genistein derivatives on the apoptosis of endothelial cells was analyzed by PI staining. influences. Results: Among all the derivatives, 7-difluoromethylene-5, 4-dimethoxy-genistein had the highest anti-vascular endothelial oxidative stress injury activity, and its effect was stronger than that of the genistein. Conclusion: The 7-difluoromethylene-5, 4-dimethoxy-genistein obtained by screening has stronger anti-vascular endothelial oxidative stress injury, which has the value of further research.
