目的探讨B7-H4在人喉癌组织中的分布以及该分子在人喉癌中表达的意义。方法免疫组化法检测人喉癌组织B7-H4、细胞增殖及上皮间质转化(epithelial mesenchymal transition,EMT)相关标志物分子的表达;免疫双荧光法检测B7-H4与EMT标志物分子的共表达情况。结果免疫组化结果显示检测62例患者中47例B7-H4阳性,阳性率75.81%;B7-H4表达在人喉癌细胞及喉癌组织浸润性淋巴细胞上,主要为胞浆及胞膜表达;此外,人喉癌组织细胞高表达细胞增殖标志物PCNA及EMT标志物如p-Smad2/3、p-Smad3、Vimentin、Snail;免疫双荧光结果显示p-Smad3、Snail、Vimentin与B7-H4共同表达于人喉癌组织细胞。结论 B7-H4广泛表达于人喉癌细胞及组织浸润性淋巴细胞,并与EMT标志物共表达,可能通过促进EMT进程导致喉癌的侵袭及转移,提示其可作为喉癌治疗的一个新靶点。
Objective To investigate the distribution of B7-H4 in human laryngeal carcinoma and its significance in human laryngeal carcinoma. Methods Immunohistochemistry was used to detect the expression of markers related to B7-H4, cell proliferation and epithelial mesenchymal transition (EMT) in human laryngeal carcinoma. Immunofluorescence staining was used to detect the expression of B7-H4 and EMT marker molecules Express the situation. Results The results of immunohistochemistry showed that the positive rate of B7-H4 in 47 cases was 75.81% in 62 cases. The expression of B7-H4 in human laryngeal carcinoma and laryngeal carcinoma infiltrating lymphocytes was mainly cytoplasm and membrane In addition, PCNA and EMT markers, such as p-Smad2 / 3, p-Smad3, Vimentin and Snail, were highly expressed in human laryngeal carcinoma cells. Immunofluorescence double staining showed that p-Smad3, Snail, Vimentin and B7-H4 Co-expression in human laryngeal cancer cells. Conclusions B7-H4 is widely expressed in human laryngeal cancer cells and infiltrating lymphocytes and co-expressed with EMT markers, which may be the new target for the treatment of laryngeal carcinoma by promoting the invasion and metastasis of laryngeal carcinoma by promoting the progression of EMT point.
