目的研究D20S85和D2S441位点的遗传多态性及其法医学应用价值。方法应用聚合酶链式反应、聚丙烯酰胺凝胶电泳及银染显带技术对温州地区212名汉族无关个体的D20S85和D2S441位点进行分型,并检验其基因型频率分布是否符合Hardy-Weinberg平衡定律。结果D20S85观察到9个等位基因及32种基因型,D2S441观察到10个等位基因及36种基因型,多态性分布符合Hardy-Weinberg平衡。两基因座的杂合度(H)分别为0.750、0.731,多态性信息量(PIC)为0.75、0.79。结论该研究所得到的等位基因频率数据可为温州汉族人群法医个体识别、亲子鉴定及遗传学研究提供更多依据。
Objective To study the genetic polymorphisms of D20S85 and D2S441 loci and their forensic value. Methods The D20S85 and D2S441 loci in 212 Han unrelated individuals in Wenzhou area were genotyped by polymerase chain reaction, polyacrylamide gel electrophoresis and silver staining with banding technique. The genotype frequency distributions were tested according to Hardy-Weinberg The law of balance Results Nine alleles and 32 genotypes were observed in D20S85. Ten alleles and 36 genotypes were observed in D2S441. The distribution of polymorphisms was consistent with Hardy-Weinberg equilibrium. The heterozygosities (H) of the two loci were 0.750 and 0.731, respectively, and the PICs were 0.75 and 0.79 respectively. Conclusion The allele frequency data obtained from this study may provide more evidences for forensic identification, paternity testing and genetics research in Wenzhou Han population.
