采集安徽庐江水稻条纹叶枯病感病稻株,提取病叶总RNA。设计特异性引物进行RT-PCR扩增,获得水稻条纹病毒安徽分离物(RSV-AH)的RNA3部分片段,克隆并测序。序列分析表明,该片段全长1 051个核苷酸,其中包含的CP基因全长969个核苷酸,编码322个氨基酸。将RSV安徽分离物CP基因(AH-CP)与其它来源的RSVCP基因进行核甘酸序列相似性比较,结果表明,安徽庐江的RSVCP基因与江苏洪泽的RSVCP基因序列相似性最高,达99.3%。进一步构建RSVCP基因系统关系树,发现安徽庐江的RSVCP基因同样与江苏洪泽的RSVCP基因亲缘关系最近。
The rice plants infected with rice stripe virus were collected in Lujiang of Anhui province, and the total RNA of diseased leaves was extracted. The specific primers were designed and amplified by RT-PCR. The RNA3 fragment of RSV-AH was obtained and cloned and sequenced. Sequence analysis showed that the fragment was 1 051 nucleotides in length and contained 969 nucleotides in length and encoded 322 amino acids. The results of nucleotide sequence similarity analysis between RSVCP gene of RSV Anhui isolate and RSVCP gene from other sources showed that RSVCP gene in Lujiang of Anhui Province had the highest similarity with RSVCP gene of Hongze in Jiangsu Province, reaching 99.3%. Further construction of the RSVCP gene phylogenetic tree found that the RSVCP gene in Lujiang, Anhui Province was also the most closely related to the RSVCP gene in Hongze, Jiangsu Province.
