目的探讨凋亡蛋白Bad、Noxa在胃癌组织中表达及其与细胞凋亡的关系。方法 40例胃腺癌患者手术切除的胃癌组织(胃癌组)及癌旁组织(癌旁组),采用免疫组织化学法检测2组Bad、Noxa蛋白表达情况,采用TUNEL法检测细胞凋亡情况。结果胃癌组Bad和Noxa蛋白阳性表达率(75%和65%)明显低于癌旁组(95%和75%)(P<0.05);胃癌组Bad蛋白阳性表达在胃癌组织分化程度上比较差异有统计学意义(P<0.05),Noxa蛋白阳性表达在胃癌组织分化程度、分期、淋巴结转移及远处转移上比较差异有统计学意义(P<0.05);胃癌细胞凋亡指数在胃癌细胞分化程度、淋巴结转移、远处转移及临床分期上比较差异有统计学意义(P<0.05);胃癌组Bad、Noxa表达阴性者胃癌细胞凋亡指数均明显低于表达阳性者(P<0.05)。结论 Bad、Noxa蛋白可能是抑制胃癌发生、发展的重要因素,对胃癌分化程度及治疗效果的评估有一定价值。
Objective To investigate the expression of apoptosis proteins Bad and Noxa in gastric carcinoma and their relationship with apoptosis. Methods 40 cases of gastric adenocarcinoma (gastric cancer group) and adjacent normal tissue (paracancer group) were resected. The expressions of Bad and Noxa protein in the two groups were detected by immunohistochemistry. The apoptosis was detected by TUNEL method. Results The positive expression rate of Bad and Noxa in gastric cancer group was significantly lower than that in adjacent non-cancerous group (95% and 75%, P <0.05). The positive expression of Bad protein in gastric cancer group was significantly lower than that in gastric cancer group (P <0.05). There was significant difference in the expression of Noxa protein in the degree of differentiation, staging, lymph node metastasis and distant metastasis (P <0.05). The apoptosis index of gastric cancer cells was significantly higher in gastric cancer cells Lymph node metastasis, distant metastasis and clinical stage (P <0.05). The apoptosis index of gastric cancer cells in gastric cancer group with negative expression of Bad and Noxa were significantly lower than those with positive expression (P <0.05). Conclusions Bad and Noxa proteins may play an important role in inhibiting the occurrence and development of gastric cancer, and have some value in evaluating the differentiation degree and therapeutic effect of gastric cancer.
