[目的]建立高效液相色谱法测定指甲油中苏丹红Ⅱ、苏丹红Ⅳ含量的方法。[方法]采用HPLC法对指甲油中苏丹红Ⅱ、苏丹红Ⅳ进行含量测定。色谱条件为:C18柱(3.5μm,4.6 mm×150 mm),柱温30℃,流动相为乙腈-缓冲溶液梯度洗脱,流速1.0 ml/min,检测波长484 nm及514 nm切换。[结果]苏丹红Ⅱ、苏丹红Ⅳ色谱分离良好,浓度与峰面积呈良好线性关系,线性范围分别为苏丹红Ⅱ0~50.26μg/ml,r=0.999 8(n=6);苏丹红Ⅳ0~49.78μg/ml,r=0.999 9(n=6)。平均加样回收率分别为95.9%(n=9,RSD=2.87%)、87.7%(n=9,RSD=0.4%)。[结论]该方法测定了5批指甲油中苏丹红Ⅱ、苏丹红Ⅳ的含量,分离度好、快速、简便、重现性好。
[Objective] To establish a method for the determination of Sudan Ⅱ and Sudan Ⅳ in nail polish by high performance liquid chromatography. [Method] The contents of Sudan Ⅱ and Sudan Ⅳ in nail polish were determined by HPLC. The chromatographic conditions were: C18 column (3.5 μm, 4.6 mm × 150 mm) with a column temperature of 30 ° C. The mobile phase was acetonitrile-buffered gradient elution at a flow rate of 1.0 ml / min and switched at 484 nm and 514 nm. [Results] The chromatograms of Sudan Ⅱ and Sudan Ⅳ were well separated and the linear relationship was found between the concentration and the peak area. The linear ranges were Sudan Ⅱ 0 ~ 50.26 μg / ml, r = 0.999 8 (n = 6) 49.78 μg / ml, r = 0.999 9 (n = 6). The average recoveries were 95.9% (n = 9, RSD = 2.87%) and 87.7% (n = 9, RSD = 0.4%), respectively. [Conclusion] The method was used to determine the content of Sudan Ⅱ and Sudan Ⅳ in 5 batches of nail polish. The method has good resolution, rapidity, simplicity and good reproducibility.
