目的:观察柴芩通淋片对慢性非细菌性前列腺炎大鼠模型的作用,探讨其发挥作用的可能机制。方法:制作慢性非细菌性前列腺炎大鼠模型,分组造模14 d后给予柴芩通淋片混悬液灌胃3周,空白对照组及模型组给予相应剂量的生理盐水灌胃,灌胃结束后取前列腺组织及前列腺液。前列腺组织石蜡包埋切片行HE染色,观察前列腺组织病变程度,免疫组化法观察TNF-α、IL-10阳性表达细胞数;前列腺液白细胞计数评级。结果:大体观察显示模型组大鼠前列腺显著充血和水肿,镜下HE染色显示实质内腺泡萎缩、腺腔变小,间质及腺腔内大量单个核细胞浸润,结缔组织增生,呈现明显的慢性炎症表现;前列腺液白细胞数量增多。用药后柴芩通淋片高、中、低剂量组大鼠前列腺炎性病变及纤维化程度得到较好的改善,前列腺液白细胞数量显著减少。模型组大鼠前列腺组织中TNF-α阳性细胞数显著增多,IL-10阳性细胞数显著下降。柴芩通淋片高、中、低剂量组大鼠TNF-α阳性表达细胞数低于模型组,柴芩通淋片高剂量组IL-10阳性细胞数高于模型组。结论:柴芩通淋片能够显著改善慢性非细菌性前列腺炎模型大鼠的炎症病理改变,其作用机制可能通过降低促炎性细胞因子TNF-α的表达及促进抗炎性细胞因子IL-10表达,与调节前列腺组织炎症反应有关。
Objective: To observe the effect of Chaiqin Tongling Tablet on chronic nonbacterial prostatitis rat model and to explore its possible mechanism. Methods: Chronic nonbacterial prostatitis rat model was made. The rats were given Chaiqintonglei suspension for 3 weeks after model grouping, and the blank control group and model group were given the corresponding dose of normal saline, After taking prostatic tissue and prostatic fluid. Paraffin embedded sections of prostate tissue were stained with HE to observe the degree of prostatic tissue lesions. Immunohistochemistry was used to observe the number of TNF-α and IL-10 positive cells; Prostatic fluid leukocyte count was graded. Results: The gross observation showed that the rats in the model group had significant hyperemia and edema in the prostatic glands. Microscopic HE staining showed that the acinar atrophy, the glandular cavity became smaller, the infiltration of numerous mononuclear cells and connective tissue in the interstitium and glandular cavity showed obvious Chronic inflammatory manifestations; Prostatic fluid leukocytes increased. After administration of Chaiqin Tongling Table, rats with high, medium and low doses of prostatitis had better inflammatory lesions and fibrosis, and the number of leukocytes in prostatic fluid was significantly reduced. The number of TNF-α-positive cells and the number of IL-10 positive cells in the model group were significantly decreased. The number of TNF-α-positive cells in Chaiqin Tongling Tablets with high, medium and low dose groups was lower than that in model group, and the number of IL-10 positive cells in Chaiqintongling high-dose group was higher than that in model group. Conclusion: Chaiqin Tongling Tablets can significantly improve the pathological changes of inflammation in chronic nonbacterial prostatitis model rats, and its mechanism may be through reducing the expression of pro-inflammatory cytokines TNF-α and promoting the anti-inflammatory cytokines IL-10 Expression, and regulation of prostate tissue inflammatory response.
