目的 探讨加铬重组复合纤维 (AFC- Cr)降低实验性非胰岛素依赖型糖尿病(NIDDM)大鼠胰岛素抵抗 (IR)的作用。方法 体重 2 50 g左右的雄性 Wistar大鼠 33只 ,随机选7只饲以基础饲料为 A组 (正常对照组 ) ,余 2 6只尾静脉注射 (2 5mg/ kg bw)链脲霉素 (STZ)饲以高糖、高脂及 1 %胆固醇饲料诱导成类似 NIDDM大鼠模型后 ,按体重和胰岛素敏感指数 (ISI)均衡的原则分为三组 :B组 (糖尿病对照组 )、C组 (糖尿病干预组 )、D组 (糖尿病治疗组 )。观察 AFC- Cr对大鼠糖、脂代谢、胰岛素敏感性及肝糖原等的影响。结果 AFC- Cr不影响摄食量 ,可降低体增重和肝体比 ;与 B组相比 ,明显改善糖耐量 ,降低血清果糖胺 ,降低胰岛素水平 ,提高 ISI;饲以AFC- Cr大鼠的血清总胆固醇、甘油三酯、低密度脂蛋白胆固醇及脂质过氧化物含量明显降低 ,高密度脂蛋白胆固醇轻度升高 ;与 NIDDM模型组相比 ,饲以 AFC- Cr大鼠的肝脂肪变性明显减轻 ,肝糖原合成增加。结论 AFC- Cr具有降低实验性 NIDDM大鼠血糖、血脂、果糖胺 ,减少体内脂质过氧化物等 ,减轻 IR的综合作用。
Objective To investigate the effect of AFC-Cr on insulin resistance (IR) in experimental non-insulin-dependent diabetes mellitus (NIDDM) rats. Methods Thirty-three male Wistar rats with a body weight of about 2 50 g were randomly selected to receive basic diet for group A (normal control group) and other 26 piglets (25 mg / kg bw) for streptozotocin STZ) were fed with high glucose, high fat and 1% cholesterol diet into NIDDM rats. The rats were divided into three groups according to the principle of body weight and insulin sensitivity index (ISI): group B (diabetic control group), group C (Diabetes intervention group), D group (diabetes treatment group). The effects of AFC-Cr on glucose, lipid metabolism, insulin sensitivity and glycogen in rats were observed. Results AFC-Cr did not affect the food intake and decreased body weight gain and liver body ratio. Compared with group B, AFC-Cr significantly improved glucose tolerance, lowered serum fructosamine, decreased insulin and increased ISI. Serum total cholesterol, triglyceride, low density lipoprotein cholesterol and lipid peroxidation decreased significantly, high density lipoprotein cholesterol increased slightly; compared with NIDDM model group, fed with AFC-Cr rat liver fat Degeneration significantly reduced, increased glycogen synthesis. Conclusion AFC-Cr can reduce blood glucose, blood lipids, fructosamine, reduce lipid peroxides in vivo and reduce IR in experimental NIDDM rats.
