目的:通过检测灵芝孢子油对人肺腺癌细胞SPC-A1增殖、凋亡与miR-21及其靶基因表达的影响,探讨灵芝孢子油的抗肿瘤机制。方法:灵芝孢子油处理人肺腺癌细胞SPC-A1 24 h以及48 h后,采用CCK-8法检测其对细胞增殖的影响,倒置相差显微镜观察细胞形态学变化,流式细胞术检测细胞凋亡,Real-time PCR检测miR-21及其靶基因PTEN和PDCD4的表达。结果:灵芝孢子油剂量、时间依耐性地抑制人肺腺癌SPC-A1细胞的增殖;当灵芝孢子油体积分数达到0.2%时,细胞形态发生明显变化;Annexin-V/PI双染检测提示,灵芝孢子油在低浓度下就可促进细胞凋亡;灵芝孢子油显著下调SPC-A1细胞中miR-21的表达并相应上调PTEN以及PDCD4基因的表达。结论:灵芝孢子油能显著抑制人肺腺癌细胞SPC-A1细胞增殖,引起细胞形态学改变。通过下调miR-21的表达,上调抑癌基因PTEN以及PDCD4的表达,从而促进肺癌细胞凋亡,达到抗肿瘤的作用。
Objective: To investigate the effects of Ganoderma lucidum spore oil on the proliferation and apoptosis of human lung adenocarcinoma cell line SPC-A1 and the expression of miR-21 and its target genes, and to explore the antitumor mechanism of Ganoderma spore oil. METHODS: Human lung adenocarcinoma cell line SPC-A1 was treated with Ganoderma lucidum spores oil for 24 h and 48 h, respectively. The cell proliferation was measured by CCK-8 assay. Real-time PCR was used to detect the expression of miR-21 and its target genes PTEN and PDCD4. Results: Ganoderma lucidum spore oil dose and time dependently inhibited the proliferation of human lung adenocarcinoma SPC-A1 cells. When the volume of Ganoderma lucidum spores oil reached 0.2%, the morphology of cells changed obviously. The results of Annexin-V / Ganoderma lucidum spores oil can promote apoptosis at low concentration; Ganoderma spore oil significantly down-regulates the expression of miR-21 in SPC-A1 cells and up-regulates PTEN and PDCD4 gene expression. Conclusion: Ganoderma lucidum spores oil can significantly inhibit the proliferation of human lung adenocarcinoma cells SPC-A1 cells and cause morphological changes. By down-regulating the expression of miR-21 and up-regulating the expression of tumor suppressor gene PTEN and PDCD4, so as to promote the apoptosis of lung cancer cells and achieve the anti-tumor effect.
