一种新的转录延伸因子Elongin能催化哺乳动物细胞中RNA pclⅡ的转录延伸速度。El-ogin复合体是由分子量分别为110、18、和15KD的A、B、C三种亚基组成的异源三聚体。VHL蛋白对Elongin的转录延伸作用有负调控作用,VHL蛋白由VHL基因(Von Hippel-Lindau tumor suppressor gene)所编码。VHL基因的突变会引起VHL型肿瘤,一种遗传性癌症综合征。VHL蛋白能与Elongin的B,C亚基紧密结合,从而抑制Elongin复合体的转录延伸活力,这可能是VHL型肿瘤抑制的基础。这些发现表明,VHL蛋白在Elongin的转录调控中起着一定的作用。
A novel transcription elongation factor Elongin catalyzes the transcription elongation rate of RNA pclII in mammalian cells. The El-ogin complex is a heterotrimer consisting of three subunits A, B, and C with molecular weights of 110, 18, and 15 KD, respectively. The VHL protein negatively regulates the transcriptional elongation of Elongin, which is encoded by the VHL gene (Von Hippel-Lindau tumor suppressor gene). Mutations in the VHL gene cause VHL tumors, a hereditary cancer syndrome. VHL protein binds closely to the B and C subunits of Elongin, thereby inhibiting the transcriptional elongation of the Elongin complex, which may be the basis for VHL tumor suppression. These findings indicate that VHL plays a role in the transcriptional regulation of Elongin.
