目的观察血红素氧合酶-1(HO-1)及基质金属蛋白酶-9(MMP-9)蛋白在兔主动脉粥样硬化病变的表达及普罗布考对HO-1、MMP-9表达的影响。方法16只新西兰大白兔建立动脉粥样硬化症模型后,随机加喂普罗布考(n=8,普罗布考组)或淀粉(n=8,淀粉组)6周。检测两组兔以及正常新西兰大白兔(n=8,正常对照组)血脂、体重和血清丙二醛含量。采用免疫组化法测定主动脉HO-1以及MMP-9蛋白的表达。结果与淀粉组相比,普罗布考组动脉粥样斑块缩小,HO-1在斑块内及动脉平滑肌内表达增多,MMP-9在斑块内表达减少,血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度胆固醇(HDL-C)和丙二醛明显降低(P均<0.05)。结论普罗布考在降低胆固醇的同时,还可能通过上调HO-1、下调MMP-9发挥作用,这可能是其抗动脉粥样硬化的重要途径。
Objective To observe the expression of heme oxygenase-1 (HO-1) and matrix metalloproteinase-9 (MMP-9) in aortic atherosclerosis and the effects of probucol on HO-1 and MMP- influences. Methods Sixteen New Zealand rabbits were randomly divided into four groups: probucol (n = 8, probucol) or starch (n = 8, starch) for 6 weeks. Blood lipid, body weight and serum MDA level were detected in two groups of rabbits and normal New Zealand white rabbits (n = 8, normal control group). Immunohistochemistry was used to detect the expression of HO-1 and MMP-9 in the aorta. Results Compared with the starch group, the atherosclerotic plaque in probucol reduced, the expression of HO-1 in plaque and arterial smooth muscle increased, the expression of MMP-9 in plaque decreased, the serum total cholesterol (TC), low LDL-C, HDL-C and MDA decreased significantly (all P <0.05). Conclusions Probucol might be an important anti-atherosclerosis mechanism by lowering HO-1 and down-regulating MMP-9 while lowering cholesterol.
