目的探讨肿瘤坏死因子(TNF)-α和肿瘤坏死因子受体(TNFR)1在妊娠期糖尿病大鼠子宫内膜中的表达变化及意义。方法取30只SD孕鼠,随机分为正常妊娠组和妊娠期糖尿病组(n=15),妊娠期糖尿病组一次性腹腔注射45mg·kg-1链脲佐菌素(STZ),正常妊娠对照组注射等量枸橼酸缓冲液;妊娠20d,检测空腹血糖(FBG)、空腹胰岛素(FINS)并计算胰岛素抵抗指数(HOMA-IR),同时,采用Western blotting检测子宫内膜TNF-α和TN-FR1的蛋白表达,分析HOMA-IR与TNF-α和TNFR1表达水平的相关性。结果 (1)妊娠期糖尿病大鼠FBG、FINS及HOMA-IR均显著高于正常妊娠组(P﹤0.01);(2)TNF-α和TNFR1在妊娠期糖尿病大鼠子宫内膜中的蛋白表达量均明显高于正常组(P﹤0.001);(3)子宫内膜TNF-α和TNFR1的蛋白表达水平与HOMA-IR呈正相关(r=0.684,P﹤0.01;r=0.579,P﹤0.05)。结论子宫内膜TNF-α和TNFR1蛋白的过表达可能是妊娠期糖尿病胰岛素抵抗的重要机制之一。
Objective To investigate the expression and significance of tumor necrosis factor (TNF) -α and tumor necrosis factor receptor (TNFR) 1 in endometrium of gestational diabetic rats. Methods Twenty SD pregnant rats were randomly divided into normal pregnancy group and gestational diabetes group (n = 15). Gestational diabetes mellitus group received intraperitoneal injection of streptozotocin (STZ) 45 mg · kg-1 and control group The rats were injected with the same amount of citrate buffer. Pregnancy 20 days, fasting blood glucose (FBG) and fasting insulin (FINS) were measured and insulin resistance index (HOMA-IR) was calculated. Meanwhile, the levels of TNF- -FR1 protein expression, analysis of HOMA-IR and TNF-α and TNFR1 expression levels. Results (1) FBG, FINS and HOMA-IR of gestational diabetic rats were significantly higher than those of normal pregnant rats (P <0.01). (2) The protein expression of TNF-α and TNFR1 in endometrium of gestational diabetic rats (P <0.001). (3) The expression of TNF-α and TNFR1 in endometrium was positively correlated with HOMA-IR (r = 0.684, P <0.01; r = 0.579, P <0.05) ). Conclusion Overexpression of TNF-α and TNFR1 in endometrium may be one of the important mechanisms of insulin resistance in gestational diabetes mellitus.
