目的:研究芭蕉根的化学成分及其生物活性。方法:在DPPH抗氧化和α-葡萄糖苷酶抑制活性指导下对芭蕉根提取物进行活性追踪,对高活性的醋酸乙酯部位采用各种色谱法分离,运用多种波谱技术鉴定结构;并对得到的化合物进行DPPH自由基清除活性、糖苷酶抑制作用和抗金黄色葡萄球菌(SA)、耐甲氧西林的金黄色葡萄球菌(MRSA)和β-内酰胺酶阳性的金黄色葡萄球菌(ESBLs)活性筛选。结果:从芭蕉根醋酸乙酯部位分离得到5个化合物:2′,3,4′-三羟基黄酮(1),3,3′-bis-hydroxyanigorufone(2),irenolone(3),2,4-dihydroxy-9-(4′-hydroxyphenyl)-phenalenone(4)和3,4-二羟基苯甲醛(5)。化合物1(IC508.61 mg.L-1),3(IC5019.55 mg.L-1)和5(IC501.1 mg.L-1)具有体外抗氧化活性;2(IC5024.15 mg.L-1)和4(IC502.81 mg.L-1)具有体外α-葡萄糖苷酶抑制活性;化合物4对SA,MRSA和ESBLs均有很强的抑制作用,最低抑菌浓度(MIC)分别为0.078,0.313,0.039μg/disc。结论:化合物1~5为首次从该植物中分离得到,5为首次从该属中分离得到;除化合物5的抗氧化活性已经报道过外,其他化合物的活性均为首次报道。
Objective: To study the chemical constituents and biological activities of plantain. Methods: The activity of banana root was traced under the guidance of DPPH antioxidant and α-glucosidase inhibitory activity. The chromatographic method was used to separate the highly active fraction of ethyl acetate. The structure was identified by using a variety of spectroscopic techniques. The obtained compounds were subjected to DPPH radical scavenging activity, glycosidase inhibition and anti-Staphylococcus aureus (SA), methicillin-resistant Staphylococcus aureus (MRSA) and β-lactamase-positive Staphylococcus aureus ) Activity screening. Results: Five compounds were isolated from the ethyl acetate fraction of banana root: 2 ’, 3,4’-trihydroxyflavone (1), 3,3’-bis-hydroxyanigorufone (2), irenolone -dihydroxy-9- (4’-hydroxyphenyl) -phenalenone (4) and 3,4-dihydroxybenzaldehyde (5). In vitro antioxidant activity of compounds 1 (IC508.61 mg.L-1), 3 (IC5019.55 mg.L-1) and 5 (IC501.1 mg.L-1); 2 (IC5024.15 mg.L 1) and 4 (IC502.81 mg.L-1) had inhibitory activity on α-glucosidase in vitro. Compound 4 showed a strong inhibitory effect on SA, MRSA and ESBLs. The MICs of compound 4 were 0.078, 0.313, 0.039 μg / disc. CONCLUSION: Compounds 1 ~ 5 were isolated from this plant for the first time. 5 was isolated from this genus for the first time. The activity of all other compounds was reported for the first time, except that the antioxidant activity of compound 5 was reported.
