目的 研究大鼠舌癌及癌前病变组织中的端粒酶活性 ,探讨其在口腔癌诊断与防治方面的应用前景。方法 采用PCR TRAP银染与ELISA技术分别对 6 9例 4NQO诱发大鼠舌癌变过程中端粒酶活性变化进行定性和定量检测。结果 两种方法检测端粒酶活性有很好的一致性。随口腔癌变进展 ,端粒酶阳性率递增 ,正常粘膜、单纯性增生、轻中度异常增生、重度异常增生、原位癌、鳞癌的阳性率分别为 0 0 %、12 5 %、2 5 0 %、6 6 7%、72 7%和 93 8%。结论 端粒酶激活是口腔癌形成的重要环节 ,检测癌前病变组织中端粒酶活性可预测癌变潜能。端粒酶为口腔癌防治研究提供了理想靶点
Objective To study the telomerase activity in rat tongue and precancerous lesions and to explore its potential application in the diagnosis and prevention of oral cancer. Methods PCR TRAP silver staining and ELISA were used to detect the changes of telomerase activity in the tongue carcinogenesis of 6 9 rats induced by 4NQO respectively. Results Both methods showed good agreement with telomerase activity. With the development of oral cancer, the positive rate of telomerase increased gradually, the positive rates of normal mucosa, simple hyperplasia, mild to moderate dysplasia, severe dysplasia, carcinoma in situ and squamous cell carcinoma were 0 0%, 125%, 25% 0%, 6 6 7%, 72 7% and 93 8%. Conclusion Telomerase activation is an important step in the development of oral cancer. Detection of telomerase activity in precancerous lesions predicts cancer potential. Telomerase provides an ideal target for the prevention and treatment of oral cancer
