目的 了解乳腺癌细胞系NFκB家族蛋白的表达及其与化疗耐药的关系。方法 用免疫印记法分别检测了14个乳腺癌细胞系的P65,P50及IκB-α的表达,同时对10个细胞系进行MTT药物敏感实验。结果 IκB-α主要在胞浆内表达,虽然P65和P50在胞浆、胞核均有表达,但无论在胞浆还是在胞核,P50的表达均占明显优势,其在胞核的表达高达63%。P50胞核表达阳性者IC50值明显高于胞核阴性者,差异有显著意义。结论 1)乳腺癌细胞核P50表达可能预示化疗耐药,故可作为判断预后的一个新指标;2)NFκB家族蛋白中,P50的表达在乳腺癌 中更常见,且在化疗耐药性的产生中可能起着更重要的作用。
Objective To investigate the expression of NFκB family of breast cancer cell lines and its relationship with chemoresistance. Methods The expression of P65, P50 and IκB-α in 14 breast cancer cell lines were detected by Western blotting, and MTT drug sensitivity test was performed in 10 cell lines. Results The expression of IκB-α was mainly in the cytoplasm. Although P65 and P50 were both expressed in cytoplasm and nucleus, the expression of P50 in both cytoplasm and nucleus showed obvious advantages. The expression of IκB- 63%. The IC50 of P50 positive cells was significantly higher than that of negative ones, the difference was significant. Conclusions1) The expression of P50 in breast cancer cell nucleus may predict chemoresistance, which may be used as a new index to judge the prognosis. 2) The expression of P50 in NFκB family protein is more common in breast cancer, and in the generation of chemoresistance May play a more important role.
