目的探讨结肠癌组织中LIN28B的表达及其临床意义。方法选择美国Biomax的结肠癌及正常组织组合微阵列芯片及2003年1月至2004年12月南方医科大学南方医院的149例手术切除结肠癌石蜡标本作为研究对象。用免疫组化法检测组织芯片和石蜡切片中LIN28B蛋白表达并评分。采用χ2检验比较不同LIN28B表达水平和结肠癌病理性特点包括组织分级、恶性肿瘤(tumor,node,metastasis,TNM)分期及淋巴/远处转移之间的关系。结果 LIN28B在结肠癌组织中相比于正常结肠组织高表达,其高表达与肿瘤的低分化、TNMⅢ-Ⅳ期、结肠癌局部淋巴结转移及远处转移有关。多因素Logistic回归分析显示TNM分期是LIN28B高表达的独立危险因素。结论 LIN28B表达与结肠癌病理特征密切相关,有望作为判断结肠癌患者预后的指标。
Objective To investigate the expression of LIN28B in colon cancer and its clinical significance. METHODS: Colon cancer and normal tissue microarray chips from US Biomax and 149 cases of resected colon cancer paraffin specimens from Nanfang Hospital of Southern Medical University from January 2003 to December 2004 were selected as research objects. The expression of LIN28B protein in tissue and paraffin sections was detected by immunohistochemistry and scored. Chi-square test was used to compare the expression of different LIN28B and the pathological features of colon cancer, including the histological grade, the relationship between the stage of tumor, node, metastasis (TNM) and lymphoid / distant metastasis. Results The high expression of LIN28B in colorectal cancer tissue was related to the poor differentiation of tumor, TNMⅢ-Ⅳ stage, local lymph node metastasis and distant metastasis of colon cancer. Multivariate Logistic regression analysis showed that TNM staging was an independent risk factor for the high expression of LIN28B. Conclusion The expression of LIN28B is closely related to the pathological features of colon cancer, which is expected to be an index to evaluate the prognosis of patients with colon cancer.
