[目的]对早期和晚期初诊鼻咽癌患者肿瘤组织在蛋白质组水平上的表达差异进行比较研究,识别和鉴定差异蛋白,并对其功能进行初步了解。[方法]按照临床分期将所收集鼻咽癌组织分为早期患者组(I、II期)和中晚期患者组(III、IV期),分别提取两组组织总蛋白,进行双向凝胶电泳(two-dimensional difference gel electrophoresis)分离,利用UMAX PowerLook1100型多功能透射扫描仪不同波长的激光扫描后,获取凝胶图像,应用PDQuest7.1.0软件包进行图像分析,切取凝胶中差异表达≥1.5倍(P≤0.001)的蛋白质斑点进行胶内酶解,应用基质辅助激光解吸/电离-飞行时间质谱仪(MALDI-TOF/MS)进行蛋白质的肽指纹图谱鉴定。[结果]翻译起始因子亚基eIF3K和eIF5A在中晚期鼻咽癌中表达丰度升高。[结论]早期和晚期鼻咽癌组织存在表达差异的蛋白,翻译起始因子亚基eIF3K和eIF5A在早期和中晚期鼻咽癌组织中的差异将为解析鼻咽癌发生发展机制提供线索,也为筛选具体潜在价值的肿瘤化疗靶位点提供理论依据。
[Objective] To comparatively study the difference of proteomics level between early-stage and late-stage nasopharyngeal carcinoma patients, to identify and identify differential proteins, and to make a preliminary understanding of their functions. [Methods] According to the clinical stage, the collected NPC tissues were divided into the early stage (stage I and stage II) and the middle stage (stage III and stage IV), and the total tissues of the two groups were respectively extracted for two - dimensional gel electrophoresis Two-dimensional difference gel electrophoresis was used to separate the gel images. After laser scanning with different wavelengths of UMAX PowerLook 1100 multi-function transmission scanner, the gel images were obtained and analyzed by PDQuest7.1.0 software package. The differential expression in the gel was more than 1.5 times P≤0.001) were digested in-gel. The peptide fingerprinting of protein was identified by MALDI-TOF / MS. [Results] The translation initiation factor subunits eIF3K and eIF5A were up-regulated in advanced nasopharyngeal carcinoma. [Conclusion] The expression differences of early and late nasopharyngeal carcinoma tissues exist. The differences of eIF3K, eIF5K and eIF5A in early and advanced nasopharyngeal carcinoma tissues will provide clues for analyzing the mechanism of nasopharyngeal carcinoma. To screen specific potential value of tumor chemotherapy target sites provide a theoretical basis.
