目的 比较阿奇霉素、红霉素、磺胺嘧啶、大蒜素体外抗弓形虫的效果。方法 用弓形虫RH株速殖子感染培养的巨噬细胞 ,每一种药物根据预实验设立 2至 3个浓度 ,分别在速殖子侵入细胞前和侵入细胞后用不同浓度的药物处理 ,每隔 1h在倒置显微镜下观察细胞及弓形虫的形态及生长情况 ,9d后取出盖玻片 ,染色透明后 ,镜下观察。再将培养液及培养细胞注入鼠腹腔 ,观察小鼠的发病情况。结果 阿奇霉素的浓度在 2 0 μg/ml时具有抑制虫体生长的作用 ,>40 μg/ml时对巨噬细胞有明显的毒性。红霉素的浓度在 10 0 μg/ml、15 0 μg/ml时可抑制弓形虫的生长与繁殖 ,但不能完全杀死虫体 ,形成假包囊。将假包囊及培养液接种小鼠腹腔 6d后小鼠发病死亡 ,解剖小鼠后 ,可见大量的虫体。没有观察到 10 0 μg/ml、2 0 0 μg/ml的磺胺嘧啶及 2 0 μg/ml、40 μg/ml的大蒜素有抑制弓形虫生长的作用。 结论 阿奇霉素具有明显杀灭速殖子的作用。低浓度的红霉素有利于包囊形成。磺胺嘧啶、大蒜素体外无杀灭速殖子的作用。
Objective To compare the effects of azithromycin, erythromycin, sulfadiazine and allicin against Toxoplasma gondii in vitro. Methods Toxoplasma gondii RH strain tachyzoites were used to infect cultured macrophages. Each drug was set at 2 to 3 concentrations according to the pre-experiment. The tachyzoites were infused into the cells and then treated with different concentrations of drugs. The morphology and growth of cells and Toxoplasma gondii were observed under an inverted microscope at intervals of 1 hour. After 9 days, the coverslips were removed and stained transparent for microscopic observation. Then the culture medium and cultured cells were injected into the abdominal cavity of mice to observe the incidence of mice. Results Azithromycin had inhibitory effect on the growth of parasites at the concentration of 20 μg / ml. When> 40 μg / ml azithromycin was obviously toxic to macrophages. Erythromycin inhibited the growth and reproduction of Toxoplasma gondii at the concentrations of 10 μg / ml and 150 μg / ml, but did not completely kill the parasites and formed pseudocysts. The mice were sacrificed 6 days after inoculation of pseudocysts and culture medium into mice’s abdominal cavity. After the mice were dissected, a large number of parasites were observed. Sulfadiazine at a concentration of 100 μg / ml, 200 μg / ml and allicin at 20 μg / ml and 40 μg / ml were not observed to inhibit the growth of Toxoplasma gondii. Conclusion Azithromycin has a clear role in killing tachyzoites. Low concentrations of erythromycin favors cyst formation. Sulfadiazine, allicin in vitro kill tachyzoite effect.
