本文报道了用小鼠胸腺细胞MTT比色分析法检测淫羊藿多糖、猪苓多糖与羧甲基茯苓多糖及微量元素锌对正常人外周血单个核细胞产生白细胞介素2(IL_2)的影响,结果表明,淫羊藿多糖在100μg/ml和1000μg/ml剂量对人IL_2的产生有显著的增强作用,在2500μg/ml和5000μg/ml表现出抑制作用;猪苓多糖与羧甲基茯苓多糖合用在一定浓度下也有显著的增强作用;微量元素锌在0.153~765μmol/L均有显著的增强作用。并证明,各药对IL_2产生的作用并非药物的直接作用所致,淫羊藿多糖高浓度的抑制作用亦非该药的细胞毒作用所致。本文建立了用小鼠胸腺细胞做靶细胞的MTT比色分析法并与~3H—TdR掺入法进行了比较,获得较好的一致性。
In this paper, we report the effect of epimedium polysaccharide, polyporus umbellatus polysaccharide, carboxymethyl guanosyl polysaccharide, and trace element zinc on the production of interleukin 2 (IL 2) in peripheral blood mononuclear cells of normal people using MTT colorimetric assay of mouse thymocytes. The results showed that epimedium polysaccharides significantly enhanced the production of human IL 2 at doses of 100 μg/ml and 1000 μg/ml, and exhibited inhibitory effects at 2500 μg/ml and 5000 μg/ml. Polyporus umbellatus polysaccharide and carboxymethyl guanosyl polysaccharide Combined use at a certain concentration also has a significant enhancement; trace elements of zinc in the range of 0.153 ~ 765μmol / L have significant enhancement. And proved that the effect of each drug on the production of IL2 is not caused by the direct effect of the drug. The inhibitory effect of high concentration of epimedium polysaccharide is not due to the cytotoxic effect of the drug. In this study, MTT colorimetric assays using mouse thymocytes as target cells were established and compared with the ~3H-TdR incorporation method to obtain good agreement.
