用90%纯度的肝细胞增殖抑制因子(HPI)制备了效价为1∶32的兔抗鼠HPI抗血清,并检测了HPI在肝内的原位表达。结果表明,正常成年大鼠、兔和豚鼠肝细胞均表达HPI,肝小叶中央带肝细胞表达尤强,非肝细胞无HPI表达。胎鼠和新生鼠肝细胞及非肝细胞无HPI表达。肾内各处结构和细胞也无HPI表达。在肝大部切除后1~2d,肝细胞无HPI表达,4d后呈弱阳性表达并逐渐增强,至10d时达正常成年鼠表达水平。在肝癌发生中,转化肝细胞和肝癌细胞无HPI表达。研究结果提示,HPI参与肝细胞的增殖分化及转化癌变过程的调控
Rabbit anti-mouse HPI antiserum with a titer of 1:32 was prepared with 90% pure hepatocyte proliferation inhibitory factor (HPI) and the in situ expression of HPI in liver was examined. The results showed that HPI was expressed in hepatocytes of normal adult rats, rabbits and guinea pigs. The expression of hepatocytes in the hepatic lobule was stronger than that in the non-hepatocytes. HPI expression in fetal and neonatal rat hepatocytes and non-hepatocytes. There were also no HPI expression in the structures and cells in the kidney. At 1 ~ 2 days after partial hepatectomy, there was no HPI expression in hepatocytes, weakly positive expression at 4 days, and gradually increased to normal expression level at 10 days. In hepatocellular carcinoma, there is no HPI expression in transformed hepatocytes and hepatoma cells. The results suggest that HPI is involved in the proliferation and differentiation of hepatocytes and the regulation of carcinogenesis
