目的建立桑叶中芦丁含量的反相高效液相色谱法测定方法并测定不同产地桑叶中芦丁的含量。方法Hy-persil BDS C18色谱柱(4.6 mm×250 mm,5μm),水和甲醇溶剂系统线性梯度洗脱,流速1 mL.min-1,检测波长320 nm,进样量10μL。结果芦丁含量测定线性方程为Y=2.795×105X-1.041×105,r2=0.999 6,线性范围为0.203~2.436μg,平均回收率为101.1%,RSD=1.93%(n=5),不同产地桑叶芦丁含量均>0.10%,其中云南产的含量最高,为0.36%。结论实验方法稳定、可靠,为桑叶的质量鉴别研究提供了实验依据。
Objective To establish a method for the determination of rutin in mulberry leaves by RP-HPLC and determine the content of rutin in mulberry leaves from different regions. Methods Hy-persil BDS C18 column (4.6 mm×250 mm, 5 μm) was eluted with a linear gradient of water and methanol solvent system. The flow rate was 1 mL.min-1, the detection wavelength was 320 nm, and the injection volume was 10 μL. Results The linear equation for determination of rutin content was Y=2.795×105X-1.041×105, r2=0.999 6, the linear range was 0.203 to 2.436 μg, the average recovery was 101.1%, and RSD was 1.93% (n=5). The content of mulberry rutin was more than 0.10%, among which Yunnan had the highest content of 0.36%. Conclusion The experimental method is stable and reliable, providing experimental basis for the quality identification of mulberry leaves.
