[目的]明确高分子亚基Bx7OE在6个品种中的分布,研究其对小麦品质影响,帮助品质性状改良。[方法]利用已有Bx7OE特异性标记,对津强1号、辽春10号及其杂交育成的5个品种进行检测,采用SDS-PAGE方法,获得7个品种高分子量亚基分布数据,并进行品质分析。[结果]7个供试品种中,有4个品种扩增出447bp的片段,分别是津强1号、津强2号、津强5号和津强6号,说明里面含有7OE亚基。SDS-PAGE数据表明,这7个品种亚基组成为在Glu-A1位点除津强1号为2*亚基外,其余均为1亚基,在Glu-B1和Glu-D1位点均为7+8亚基和5+10亚基。通过品质数据分析,Bx7OE亚基对小麦粉质指标有明显的正面影响。[结论]STS标记特异性较好,Bx7OE亚基对改良小麦品质作用较大,可用于中国小麦品质改良。
[Objective] The research aimed to clarify the distribution of polymer subunit Bx7OE in 6 varieties and study its impact on wheat quality and help to improve the quality traits. [Method] The existing Bx7OE specific markers were used to detect the five cultivars of Jinqiang 1, Liaochun 10 and their hybrids. The SDS-PAGE method was used to obtain the distribution data of seven high molecular weight subunits Conduct quality analysis. [Result] A total of 447bp fragments were amplified from 4 cultivars, Jinqiang 1, Jinqiang 2, Jinqiang 5 and Jinqiang 6, which indicated that they contained 7OE subunits. The results of SDS-PAGE showed that the subunits of these seven cultivars were all subunit 1 at the Glu-A1 locus except for the 2 * subunit of Jinqiang 1, and both subunits at Glu-B1 and Glu-D1 7 + 8 subunits and 5 + 10 subunits. Through quality data analysis, Bx7OE subunit has a significant positive impact on wheat silty index. [Conclusion] The STS marker specificity was better. The Bx7OE subunit had a greater effect on improving wheat quality and could be used to improve the quality of Chinese wheat.
