目的:找出直、结肠癌中BAX基因突变与抗癌药耐药性之间的关系.方法:用MTT法测定17种常用抗癌药对65例肠癌的体外敏感性,并用聚合酶链反应单链构象多态分析技术检测其BAX基因突变情况,将之分成BAX基因正常组和突变组,并比较两组中抗癌药抑制率大小.结果:BAX基因正常组抗癌药抑制率大于突变组,其中有8种抗癌药两组存在显著差异.结论:当直、结肠癌中存在BAX基因突变时,抗癌药耐药性增加.
OBJECTIVE: To find out the relationship between BAX gene mutations and anticancer drug resistance in patients with colorectal cancer and colon cancer. Methods: To determine the sensitivity of 17 commonly used anticancer drugs to 65 cases of colorectal cancer in vitro and use polymerase chain The single-strand conformation polymorphism analysis technique was used to detect the mutation of BAX gene and divided into BAX gene normal group and mutation group. The inhibition rate of anticancer drugs in two groups was compared. Results: The inhibition rate of anticancer drugs in BAX gene normal group was greater than In the mutant group, there were significant differences between the 8 anticancer drugs and the two groups. Conclusion: When there are BAX gene mutations in straight and colon cancers, the anticancer drug resistance increases.
