目的研究CD82/KAI1在胶质瘤CHG-5及U251细胞株的表达及意义。方法 RT-PCR、免疫细胞化学、免疫荧光检测CHG-5及U251细胞中CD82/KAI1的表达;分别设立1:100、1:200CD82/KAI1抗体干预组和空白对照组,通过划痕实验对CD82/KAI1对细胞迁移能力的影响进行分析。结果①U251细胞中CD82/KAI1的mRNA表达高于CHG-5细胞(P<0.01);U251细胞中CD82/KAI1的蛋白水平表达高于CHG-5细胞(P<0.01)。②CD82/KAI1抗体干预组迁移细胞数量均显著高于对照组(P<0.05),且迁移细胞数量随抗体浓度升高而增加(P<0.05)。结论 CD82/KAI1在恶性胶质瘤细胞株中表达,且在U251细胞中表达较CHG-5细胞高。CD82/KAI1可抑制胶质瘤U251细胞的迁移。
Objective To study the expression and significance of CD82 / KAI1 in human glioma CHG-5 and U251 cell lines. METHODS: The expression of CD82 / KAI1 in CHG-5 and U251 cells was detected by RT-PCR, immunocytochemistry and immunofluorescence. The CD82 / KAI1 cells were treated with 1: 100,1: 200 CD82 / KAI1 antibody and blank control group respectively / KAI1 on cell migration ability analysis. Results ① The mRNA expression of CD82 / KAI1 in U251 cells was higher than that in CHG-5 cells (P <0.01). The protein level of CD82 / KAI1 in U251 cells was higher than that in CHG-5 cells (P <0.01). ② The number of migrated cells in the CD82 / KAI1-treated group was significantly higher than that in the control group (P <0.05), and the number of migrated cells increased with the increase of the antibody concentration (P <0.05). Conclusions CD82 / KAI1 is expressed in malignant glioma cell lines and expressed higher in U251 cells than in CHG-5 cells. CD82 / KAI1 inhibits glioma U251 cell migration.
