目的探讨益气温阳活血化瘀方药——心复康口服液对腹主动脉部分缩窄致压力超负荷模型大鼠心脏功能和心肌能量代谢的影响。方法将120只成年雄性SD大鼠采用随机数字表法分为3组:假手术组、腹主动脉缩窄模型组(模型组)、心复康治疗组(治疗组)。采用腹主动脉部分缩窄法制作压力超负荷大鼠模型,BL-420E+生物信号采集系统测定心功能参数,用高效液相法(HPLC)测定心肌高能磷酸盐的浓度,通过透射电镜观察大鼠心肌线粒体的形态,比较3组大鼠第6、8周心功能的各项指标(SAP、DAP、LVEDP、LVMI、+dp/dtmax、-dp/dtmax)及心肌高能磷酸盐(ATP、ADP、AMP、TAN、EC)的含量。结果①与假手术组相比,造模后第6周起,模型组大鼠左心室质量指数(LVMI)、左心室舒张末压(LVEDP)显著升高,左心室内压最大变化速率(+dp/dtmax)显著下降(P<0.01);心肌ATP、ADP、AMP含量显著降低(P<0.01);造模后第8周,上述改变更显著(P<0.01,P<0.05)。②和模型组相比,治疗组大鼠心功能指标及ATP、ADP、AMP、TAN的含量均明显改善(P<0.01)。③治疗组大鼠心功能指标及ATP、ADP、AMP、TAN的含量在8周时明显高于6周(P<0.01,P<0.05)。④治疗组大鼠心肌线粒体的受损程度较模型组轻。结论心复康口服液能明显改善慢性压力超负荷大鼠心肌能量代谢和心脏功能,并对心肌线粒体起到保护作用。
Objective To investigate the effect of Yiqiyangyanghuoxuehuayu recipe-Xinfukang oral liquid on cardiac function and myocardial energy metabolism in rats with pressure overload induced by abdominal aorta partial constriction. Methods One hundred and twenty adult male Sprague-Dawley rats were randomly divided into three groups: sham operation group, abdominal aorta constriction model group (model group), Xinfukang treatment group (treatment group). A pressure overload rat model was established by partial abdominal aortic constriction. BL-420E+ biological signal acquisition system was used to measure cardiac function parameters. The concentration of high-energy phosphate in cardiac muscle was determined by HPLC and the rats were observed by transmission electron microscopy. The morphology of myocardial mitochondria was compared between the three groups of rats at 6 and 8 weeks of cardiac function (SAP, DAP, LVEDP, LVMI, +dp/dtmax, -dp/dtmax) and myocardial high-energy phosphate (ATP, ADP, AMP, TAN, EC) content. Results 1 Compared with the sham-operated group, the left ventricular mass index (LVMI), left ventricular end-diastolic pressure (LVEDP), and the maximum rate of left intraventricular pressure (LVEDP) increased significantly in the model group from the 6th week after model establishment. Dp/dtmax decreased significantly (P<0.01); Myocardial ATP, ADP, and AMP levels decreased significantly (P<0.01); at 8 weeks after modeling, the above changes were more significant (P<0.01, P<0.05). 2 Compared with the model group, the cardiac function parameters and ATP, ADP, AMP, TAN levels in the treatment group were significantly improved (P<0.01). 3 The content of cardiac function and ATP, ADP, AMP, TAN in the treatment group was significantly higher than that in 6 weeks at 8 weeks (P<0.01, P<0.05). 4 The damage of myocardial mitochondria was lower in the treatment group than in the model group. Conclusion Xinfukang Oral Liquid can significantly improve myocardial energy metabolism and cardiac function in chronic pressure overload rats and protect myocardial mitochondria.
